Highly multiplexed quantitation of gene expression on single cells

Abstract: Highly multiplexed, single-cell technologies reveal important heterogeneity within cell populations. Recently, technologies to simultaneously measure expression of 96 (or more) genes from a single cell have been developed for immunologic monitoring. Here, we report a rigorous, optimized, quantitative methodology for using this technology. Specifically: we describe a unique primer/probe qualification method necessary for quantitative results; we show that primers do not compete in highly multiplexed amplificati… Show more

“…First, it allows enrichment of specific populations of cells in a precise and scalable manner, before transcriptional analysis. This enrichment makes the analysis of rare events feasible and establishes a clear structure for comparisons among different groups 55 . Such classification is critical for meaningful analysis of the highly variable and multiplexed data from transcriptional studies.…”

“…Such investigations require averaging the signals and statistical post processing of the data. However, it is shown that in many cases, such as gene expression, protein expression levels, and phenotypic outputs [1][2], the averaging of the signals from a population can be misleading.…”
