Endothelial dysfunction, characterized by impairment of endothelial nitric oxide synthase (eNOS) and nitric oxide (NO) bioavailability, has been implicated in diabetic cardiovascular pathogenesis. In this study, low-molecular-weight fucoidan (LMWF), which has multiple biological activities including anti-inflammatory and anti-oxidative properties, was investigated for its protective effect against endothelial dysfunction in Goto-Kakizaki type 2 diabetic rats. LMWF (50, 100, or 200 mg/kg/day) or probucol (100 mg/kg/day) were given to diabetic rats for 12 weeks. Basal blood pressure, acetylcholine-or flow-mediated relaxation of mesenteric and paw arteries, endothelium-dependent dilation of aorta, eNOS phosphorylation, and NO production were measured using laser Doppler flowmetry, force myograph, hematoxylin and eosin staining, western blot analysis, and an NO assay. We found that LMWF robustly ameliorated the basal hypertension and impairment of endothelium-dependent relaxation in the aorta, as well as mesenteric and paw arteries in diabetic rats. In addition, the reduction in eNOS phosphorylation at Ser1177, eNOS expression, and NO production because of diabetes were partially reversed by LMWF treatment. However, probucol, a lipid-modifying drug with antioxidant properties, displayed only mild effects. Moreover, LMWF induced, in a dose-dependent manner, endotheliumdependent vasodilation and eNOS phosphorylation at Ser1177 in normal aorta, and also promoted Ser1177 phosphorylation and NO synthesis in primary cultured vasoendothelial cells. Thus, these data demonstrate for the first time that fucoidan protects vasoendothelial function and reduces basal blood pressure in type 2 diabetes rats via, at least in part, preservation of eNOS function. Fucoidan is therefore a potential candidate drug for protection of endothelium in diabetic cardiovascular complications. These patients are susceptible to atherosclerosis, hypertension, and peripheral vascular disease. 1,2 It is well established that endothelium-derived nitric oxide (NO) maintains vascular homeostasis and health through vasodilation and protection of vasculature from various damaging agents. Abstract. Glycoprotein isolated from Laminaria japonica (LJGP) is known to exhibit significant cytotoxic activity against human cancer cells; however, the mechanisms of its cytoxicity are poorly understood. In this study, we investigated further possible mechanisms by which LJGP exerts its anti-cancer action in cultured human gastric carcinoma AGS cells. LJGP treatment of AGS cells resulted in inhibition of growth and induction of apoptosis in a time-and concentrationdependent manner, as determined by MTT assay, fluorescence microscopy, and flow cytometry analysis. The increase in apoptosis was associated with up-regulation of pro-apoptotic Bax expression, down-regulation of anti-apoptotic Bcl-2 and IAP family members, and activation of caspase-3 and -9. LJGP treatment markedly down-regulated the activity of telomerase and expression of human telomerase reverse transcriptase, a main determinant of telomerase enzymatic activity, with inhibition of Sp1 and c-Myc expression in a concentration-dependent manner. Furthermore, LJGP treatment also caused a progressive decrease in the expression levels of cyclooxygenase (COX)-2 without significant changes in the levels of COX-1, which was correlated with a decrease in prostaglandin E 2 synthesis. These results provide important new insights into the possible molecular mechanisms of the anti-cancer activity of LJGP.

Vi sinh vật được nuôi cấy trên đĩa thạch chứa fucoidan, chủng vi khuẩn có hoạt tính là khi vùng nuôi cấy không bị nhuộm bởi hexadecyltrimethylammonium bromide (cetavlon) 1% (w/v) mà hình thành vùng trong suốt. Trong số 44 chủng vi sinh vật được phân lập từ hải sâm, sò ốc và cầu gai, 2 chủng có khả năng sinh enzyme phân cắt fucoidan từ cả hai loại rong Sargassum mcclurei và Sargassum polycystum, 4 chủng sinh enzyme chỉ phân cắt fucoidan của một loại rong biển. Hoạt tính sinh học của fucoidan phụ thuộc vào nguồn phân lập. Fucoidan từ rong nâu thuộc họ homovà heteropolysaccharide, có thành phần chính là các gốc fucose được sulphat hóa tại vị trí C2 và C4 và liên kết với nhau bởi các liên kết (1→3) và/hoặc (1→4). Ngoài ra trong phân tử fucoidan cũng có mặt một lượng nhỏ galactose, mannose, xylose, glucose, glucuronic acid [3,8].Fucoidan có hoạt tính dược học đa dạng, tuy nhiên chúng vẫn chưa được sử dụng thành công trong việc chế tạo thuốc do khối lượng phân tử lớn và cấu trúc không rõ ràng [19]. Việc tạo ra các oligosaccharide khối lượng phân tử thấp sẽ giúp giải quyết được vấn đề này. Gần đây, cũng đã có một số nghiên cứu về sử dụng enzyme làm công cụ bẻ ngắn mạch fucoidan thành các oligosaccharide theo định hướng sử dụng trong dược học [2,9]. Enzyme phân cắt fucoidan bao gồm 2 nhóm: fucoidanase và α-L-fucoidanase. Các enzyme này cắt các liên kết glycosidic đặc hiệu trong chuỗi polysaccharide, do đó bảo tồn được các nhóm sulphate, là nhóm có vai trò quan trọng đối với hoạt tính sinh học của fucoidan [15].Cho đến nay, enzyme phân cắt fucoidan đã được tìm thấy ở một số sinh vật biển: vi khuẩn [7,10,19], nấm [16,20] và động vật thân mềm [6,18]. Tuy nhiên, fucoidanase từ các sinh vật này có hoạt tính thấp. Việc tìm kiếm các fucoidanase mới và nghiên cứu các đặc điểm động học của chúng sẽ giúp làm rõ mối quan hệ giữa cấu trúc và hoạt tính sinh học của fucoidan cũng như phát triển công nghệ sản xuất các fuco-oligosaccharide có hoạt tính sinh học quan trọng. Một trong những khó khăn chính trong nghiên cứu fucoidanase hiện nay là chưa có phương pháp đơn giản và nhạy để xác định hoạt tính xúc tác của chúng.
