Fucoidanases are endo-fucoidanases (also known as endo-fucanases) that catalyze hydrolysis of α-glycosidic linkages in fucoidans, a family of sulfated fucose-rich polysaccharides primarily found in the cell walls of brown seaweeds. Fucoidanases are promising tools for producing bioactive fucoidan oligosaccharides for a range of biomedical applications. High sulfation degree has been linked to high bioactivity of fucoidans. In this study, a novel fucoidanase, Fhf2, was identified in the genome of the aerobic, Gram-negative marine bacterium Formosa haliotis. Fhf2 was found to share sequence similarity to known endo-α(1,4)-fucoidanases (EC 3.2.1.212) from glycoside hydrolase family 107. A C-terminal deletion mutant Fhf2∆484, devoid of 484 amino acids at the C-terminus, with a molecular weight of approximately 46 kDa, was constructed and found to be more stable than the full-length Fhf2 protein. Fhf2∆484 showed endo-fucoidanase activity on fucoidans from different seaweed species including Fucus evanescens, Fucus vesiculosus, Sargassum mcclurei, and Sargassum polycystum. The highest activity was observed on fucoidan from F. evanescens. The Fhf2∆484 enzyme was active at 20–45°C and at pH 6–9 and had optimal activity at 37°C and pH 8. Additionally, Fhf2∆484 was found to be calcium-dependent. NMR analysis showed that Fhf2∆484 catalyzed hydrolysis of α(1,4) linkages between L-fucosyl moieties sulfated on C2 (similar to Fhf1 from Formosa haliotis), but Fhf2∆484 in addition released oligosaccharides containing a substantial amount of 2,4-disulfated fucose residues. The data thus suggest that the Fhf2∆484 enzyme could be a valuable candidate for producing highly sulfated oligosaccharides applicable for fucoidan bioactivity investigations.
Vibriosis in farmed animals is a serious threat to aquaculture worldwide. Using probiotics and anti-Vibrio antimicrobial substances in aquaculture systems can be a means of preventing Vibrio infections. Therefore, we aimed to characterize and compare 16 potential anti-Vibrio probiotics (Vi+) isolated from marine sponges and fish intestines collected from the Vietnam Sea, as well as an anti-Vibrio bacteriocin to fully explore their application potentials. 16S rRNA sequencing confirmed all Vi+ to be Bacillus species with different strain variants across two sample types. An obvious antimicrobial spectrum toward Gram-negative bacteria was observed from intestinal Vi+ compared to sponge-associated Vi+. The reason was the higher gene frequency of two antimicrobial compounds, non-ribosomal peptides (NRPS) and polyketide type-I (PKS-I) from intestinal Vi+ (66.7%) than sponge-associated Vi+ (14.3% and 0%, respectively). Additionally, a three-step procedure was performed to purify an anti-Vibrio bacteriocin produced by B. methylotrophicus NTBD1, including (i) solvent extraction of bacteriocin from cells, (ii) hydrophobic interaction chromatography, and (iii) reverse-phase HPLC. The bacteriocin had a molecular weight of ~2–5 kDa, was sensitive to proteolysis and thermally stable, and showed a broad antimicrobial spectrum, all of which are essential properties for promising feed additives. This study provides necessary information of the potential of probiotic Bacillus species with anti-Vibrio antimicrobial properties to study their further use in sustainable aquaculture.
k-Oligocarrageenan (OC) is an effective biostimulator and a protector against disease infections for plants. However, the effect of OC molecular weight (MW) on plant growth is not fully understood. In this work, OCs with three different MWs (42, 17 and 4 kDa) was prepared by varying the degradation reaction time using ascorbic acid as a reagent. The product structure was confirmed by Fourier-transform infrared spectroscopy (FTIR) data. The growth promotion for maize (Zea mays L.) plants was investigated by foliar spray application of the prepared OCs. Field trials were carried out in two years, 2018 and 2019. The results showed that among treatments, OC with 4 kDa exhibited the best performance in both crop growth and grain yield parameters which indicated increases compared to the control in plant height (6.9–19.9%), length of ears (12.2%), diameter of ears (9.1%), fresh grain weight (17.8%), dry grain weight (20.0%) and grain yield (21.3%). Moreover, low MW OC augmented NP uptake in the plant growth while no effect on K uptake was observed. Therefore, OC with low MWs is potentially promising to apply as a promoter to enhance yield of crops.
Introduction: M. citrifolia is known as a medical plant in Vietnam, named Noni, which contains numerous bioactive ingredients and applying good in functional food and pharmaceuticals. The notices on the change of physicochemical characteristics (color, pH, total solids, and titration acid), the content of flavonoids and polyphenol, and antioxidant activities (total, reducing power, and 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) free radical scavenging) of different extracts from Morinda citrifolia fruit at various drying methods (solar energy, microwave, and heat pump), microwave power, and temperature have not been found. Therefore, the paper focused on these things. Methods: Morinda citrifolia fruit were harvested in July 2020, dried by using different methods (solar energy, microwave, and heat pump), and soaked in 96% solvent (ethanol and aqueous) for evaluating physicochemical characteristics, flavonoids and polyphenol content, and antioxidant activities. Drying M. citrifolia was at the temperature (50, 60, and 70°C) and the microwave power (100, 200, 400, 600, and 800 W), corresponding to the heat pump method and the microwave method, respectively and evaluating the drying speed curve basing on the moisture of M. citrifolia according to the drying time. Results: Physicochemical characteristics, flavonoids content, polyphenol content, and antioxidant activities (total, reducing power, and DPPH free radical scavenging) were affected by various drying methods, different extracting solvents (p<0.05) and changed the drying time, microwave power, and temperature according to the non-linear model of level 2. The stabilization of physicochemical characteristics, flavonoids content, polyphenol content, and antioxidant activities of M. citrifolia in the drying method of microwave power was the highest, followed by the heat pump and solar energy. Ethanol solvent was more effective than aqueous solvent as extracting bioactive substances (flavonoids content and polyphenol content). Conclusion: The results will be useful for preparing the dried M. citrifolia to serve for the storage and the produce of M. citrifolia in functional food and pharmaceuticals.

Trong số 44 chủng vi sinh vật được phân lập từ hải sâm, sò ốc và cầu gai, 2 chủng có khả năng sinh enzyme phân cắt fucoidan từ cả hai loại rong Sargassum mcclurei và Sargassum polycystum, 4 chủng sinh enzyme chỉ phân cắt fucoidan của một loại rong biển. MỞ ĐẦUFucoidan là nhóm sulphat polysacarit có hoạt tính sinh học đa dạng: kháng virus [12], chống huyết khối [21], kháng viêm và kháng ung thư [1,5] . Hoạt tính sinh học của fucoidan phụ thuộc vào nguồn phân lập. Fucoidan từ rong nâu thuộc họ homovà heteropolysaccharide, có thành phần chính là các gốc fucose được sulphat hóa tại vị trí C2 và C4 và liên kết với nhau bởi các liên kết (1→3) và/hoặc (1→4). Ngoài ra trong phân tử fucoidan cũng có mặt một lượng nhỏ galactose, mannose, xylose, glucose, glucuronic acid [3,8].Fucoidan có hoạt tính dược học đa dạng, tuy nhiên chúng vẫn chưa được sử dụng thành công trong việc chế tạo thuốc do khối lượng phân tử lớn và cấu trúc không rõ ràng [19]. Việc tạo ra các oligosaccharide khối lượng phân tử thấp sẽ giúp giải quyết được vấn đề này. Gần đây, cũng đã có một số nghiên cứu về sử dụng enzyme làm công cụ bẻ ngắn mạch fucoidan thành các oligosaccharide theo định hướng sử dụng trong dược học [2,9]. Enzyme phân cắt fucoidan bao gồm 2 nhóm: fucoidanase và α-L-fucoidanase. Các enzyme này cắt các liên kết glycosidic đặc hiệu trong chuỗi polysaccharide, do đó bảo tồn được các nhóm sulphate, là nhóm có vai trò quan trọng đối với hoạt tính sinh học của fucoidan [15].Cho đến nay, enzyme phân cắt fucoidan đã được tìm thấy ở một số sinh vật biển: vi khuẩn [7,10,19], nấm [16,20] và động vật thân mềm [6,18]. Tuy nhiên, fucoidanase từ các sinh vật này có hoạt tính thấp. Việc tìm kiếm các fucoidanase mới và nghiên cứu các đặc điểm động học của chúng sẽ giúp làm rõ mối quan hệ giữa cấu trúc và hoạt tính sinh học của fucoidan cũng như phát triển công nghệ sản xuất các fuco-oligosaccharide có hoạt tính sinh học quan trọng. Một trong những khó khăn chính trong nghiên cứu fucoidanase hiện nay là chưa có phương pháp đơn giản và nhạy để xác định hoạt tính xúc tác của chúng.
