High-Throughput Transcriptome Profiling in Drug and Biomarker Discovery

Abstract: The development of new drugs is multidisciplinary and systematic work. High-throughput techniques based on "-omics" have driven the discovery of biomarkers in diseases and therapeutic targets of drugs. A transcriptome is the complete set of all RNAs transcribed by certain tissues or cells at a specific stage of development or physiological condition. Transcriptome research can demonstrate gene functions and structures from the whole level and reveal the molecular mechanism of specific biological processes in d… Show more

“…Nowadays, the rapid growth of high-throughput technologies (such as microarray and RNA-seq) has provided new approaches for gene expression profiling. A systems biology approach as a holistic method is a good choice for biomarker discovery, finding novel genes and pathways which are involved in many diseases (Yang et al, 2020).…”

“…Analysis of RNA-sequencing (RNA-seq) is an efficient technique to examine the pathological effects of disease and screening the mechanisms of drugs at tissue and/or cell level by investigating the whole transcriptional responses of host to such pathogen and drug [12]. Several studies have examined the mechanisms of probiotics on transcriptional levels by low-throughput techniques, such as quantitative polymerase chain reaction (qPCR) and/or northern blotting [13,14].…”

“…Indeed, our choice of analytical standard high-flow chromatography reduces the burden to implement proteome technologies in clinical laboratories. Second, the new high throughput capacities render proteomics applicable to drug screens, were due to a lack of sufficient throughput, so far molecular readouts were largely restricted to transcriptional profiling or metabolomics 43,44 .…”
