High-throughput analysis of fatty acid composition of plasma glycerophospholipids

Abstract: This article is available online at http://www.jlr.org ing the relationship between FA status and cardiovascular disease ( 3 ). Furthermore, the availability of n-3 long-chain polyunsaturated fatty acids (LC-PUFAs) has been related to mental development in infants ( 4 ) as well as to the attenuation of the decline of mental performance in the elderly ( 5 ). The n-6 LC-PUFA dihomo-␥ -linolenic acid and arachidonic acid and the n-3 LC-PUFA eicosapentaenoic acid are precursors of eicosanoids with different biolog… Show more

“…Lipids, another class of metabolites associated with obesity in some publications, can be determined as a molecular species or through the FA composition of single lipid classes such as glycerophopholipids [30,31] , NEFA [32] , and acylglycerols [33] , as well as that of all lipid classes (total FA; tFA) [34] . Thus, comparison of the results of different reports is challenging.…”

“…To analyze FA concentrations, EDTA plasma samples were picked and transported to the Division of Metabolic Diseases and Nutritional Medicine, Dr. von Hauner Children's Hospital, University of Munich Medical Center, in 2010. After being thawed, the analysis of plasma glycerophospholipid FA (hereafter referred to as "plasma FA") composition was performed by a sensitive and precise high-throughput method described recently (24). The average coefficient of variation was 15.7%.…”

“…Samples were centrifuged, and serum was frozen in plastic vials and stored at 280°C until analyzed. The analysis of serum glycerophospholipid fatty acid composition was performed by a sensitive and precise high-throughput method described recently (26). In brief, 100 lL serum, 100 lL internal standard (1,2-dipentadecanoyl-sn-glycero-3-phosphocholine dissolved in methanol), and 0.6 mL methanol (precooled to 5°C) were combined in glass tubes and shaken for 30 s. Samples were centrifuged and the supernatant fluid containing phospholipids and nonesterified fatty acids were transferred into additional glass tubes.…”
