High-Risk Human Papillomavirus Reduces the Expression of MicroRNA-218 in Women with Cervical Intraepithelial Neoplasia

Abstract: This study was designed to investigate whether there is a correlation between the down-regulation of microRNA-218 (miR-218) and the presence of human papillomavirus (HPV) infection in the pathogenesis of cervical cancer. The participants comprised 78 women with cervical intraepithelial neoplasia (CIN); 22 (28.2%) had CIN 1, 27 (34.6%) had CIN 2 and 29 (37.2%) had CIN 3. MiR-218 expression was determined by reverse transcriptase polymerase chain reaction and HPV genotypes in tissue specimens were identified wit… Show more

“…Notably, cervical intraepithelial neoplasia and cervical carcinoma cells with high-risk HPV infection compared to intermediate-or low-risk HPV infections also showed reduced miRNA 218 expression (21,22). Based on the current knowledge of other tumors, the downregulation of miRNA 125a-5p and miRNA 218 supposedly increases proliferation by loss of the tumor suppressive activity in JAs.…”

“…In vitro and in vivo studies demonstrated that miR-218 is down-regulated by E6 oncoprotein in both precancerous lesions and cervical cancers. In an in vivo study, Li's group [68] proposed to investigate whether there is a correlation between HPV16 infection and miR-218 down-regulation and to evaluate the relationship between miR-218 expression and CIN staging. Seventy-eight CIN cases were included in this study.…”

“…MiR-218 is remarkably down-regulated in the HPV positive cell lines, cervical lesions and cancer tissues. It may also be involved in cervical intraepithelial neoplasia (CIN) staging, due to its lower expression levels in patients with high-risk CIN than in those with low-risk CIN [30]. Decreased miR-218 expression was also connected with advanced clinical stage, lymph node metastasis, and poor prognosis in cervical cancer [31,32] and pancreatic ductal adenocarcinoma [12,33].…”
