High-Pressure Liquid Chromatographic Determination of Vitamin D3 in Resins, Oils, Dry Concentrates, and Dry Concentrates Containing Vitamin A

Abstract: A high-pressure liquid chromatographic (HPLC) procedure has been developed for the determination of vitamin D3 in resins, oils, dry concentrates, and dry concentrates containing vitamin A. Specificity of the method for vitamin D3 in the presence of isomers and other related constituents was shown by ultimate recovery of the vitamin D3 and the individual constituents and their characterization by other methods such as ultraviolet spectrophotometry. Precision and accuracy are within 2%, and as many as 20 determi… Show more

“…The solution also contained antioxidant ingredients (tertiary butylhydroquinone and citric acid) and an antifoaming ingredient (dimethylpolysiloxane). Confirmation of vitamin D 3 concentrations were performed using an HPLC technique (Hofsass et al, 1976). Dosage treatments were 1) a control 1 mL oral dosage with no supplemental vitamin D 3 , 2) 1 mL of treatment 1 but containing 40,000 IU vitamin D 3 , or 3) 1 mL of treatment 1 but containing 80,000 IU vitamin D 3 .…”

“…Very few papers described the analysis of these compounds. The isobaric impurities have been mainly studied qualitatively in the context of degradation studies or to ensure method selectivity in the determination of vitamin D3 11,[13][14][15][16][17][18] . Because it is an analogue to vitamin D, some studies have described the determination of dihydrotachysterol in biological matrices to determine the antirachitic activity, its metabolization and/or bioavailability 19,20 .…”

“…(1) This reaction was selected for use in the measurement of Fe3+ partially on the basis of studies by Hamilton et al (9,10). The authors found that the role of Fe3+ was that of a catalyst, with over 20 molecules of product produced per Fe3+ ion present.…”
