High-performance liquid chromatography–tandem mass spectrometry for identification of isoflavones and description of the biotransformation of kudzu root

Abstract: High-performance liquid chromatography-tandem mass spectrometry has been used to identify isoflavone aglycones and glycosides in kudzu root. Fourteen isoflavones were detected. Among these, six were identified by comparison with authentic standards. Tentative identifications of the other isoflavones are based on UV spectra, mass spectra of protonated and deprotonated molecules, and MS-MS data. Several are reported for the first time in kudzu root. The bioactivity and bioavailability of isoflavone aglycones are… Show more

“…Sibao et al (2007) reported puerarin, diadzin, genistein, daidzein and formononetin in the root of kudzu plant cultivated in China. Puerarin, diadzin, genistein and daidzein have been reported in kudzu root by other researchers (Zhang et al 2005;Prasain et al 2007;Chen et al 2001). In addition to the four isoflavones mentioned in the previous statement, Benlhabib et al (2004) found biochanin A in the kudzu root.…”

“…Chromatography was carried out on a Shin-pack VP-ODS column (250 mm × 4.6 mm, 5.0 m) at 25 • C. A Rheodyne 7725i sampling valve (Cotati, USA) with a sample loop of 20 L was used for sample injection. The optimum resolution was obtained by using a linear gradient elution (Lee et al, 2005;Lin et al, 2005;Zhang et al, 2005). The mobile phase consisted of (A) methanol and (B) 0.5% aqueous acetic acid using a gradient elution of 0%A and 0.1-0.5 mL/min at 0-20 min, 0-40%A and 0.5-1 mL/min at 20-35 min and 40-100%A and 1 mL/min at 35-50 min.…”

“…In the fingerprints of Puerariae lobatae and Puerariae thomsonii, 19 peaks were identified by HPLC-ESI-MS and compared with the reported MS data, 25,26) and the compounds listed in Table 1. The results suggested that the compounds were flavonoids except for Sophoraside A (peak 15).…”
