High‐performance liquid chromatography/nano‐electrospray ionization tandem mass spectrometry, two‐dimensional difference in‐gel electrophoresis and gene microarray identification of lymphatic metastasis‐associated biomarkers

Abstract: The potential biomarkers for the lymphatic metastatic process of mouse hepatocarcinoma were investigated by using two-dimensional difference in-gel electrophoresis (2D DIGE), high-performance liquid chromatography/nano-electrospray ionization tandem mass spectrometry (HPLC/nESI-MS/MS) and GeneChip. 2D DIGE was performed to screen and quantify the differentially expressed proteins between two well-established mouse hepatocarcinoma cell lines, Hca-F with 75% and Hca-P with 25% metastasis rate of lymph node poten… Show more

“…Previous proteomics work from our group, which used two-dimensional difference in gel electrophoresis (2-D DIGE) coupled high-performance liquid chromatography/HPLC-electro-spray ionisation mass spectrometry (HPLC-nESI-MS/MS) analysis, indicated that 113 proteins were differentially expressed between the two cell lines. The expression levels of annexinA7, Uchl3 and ER protein 29 were doubly validated by Western blotting analysis [2,8]. Our proteomic results of Uchl3 and ERp29 are consistent with their role in the progression of breast cancer [8,9] and protein function [10].…”

“…Therefore, annexin A7 is functionally involved in the tumour malignancy progression and is potentially associated with LNM [8]. Seventeen out of the identifi ed proteins, including Fbp2, Glud1, Crk, Ndrg1, Tkt, Anxa5, Krt2-7, Krt2-8 and Lactb2, were found to be differentially expressed, in accordance with their mRNA levels, which might be critical protein candidates associated with the LNM of hepatocarcinoma [2].…”

“…Alterations in the activity of human TKT have been reported to cause and/or accompany different pathological disorders, including the Wernicke-Korsakoff syndrome, Alzheimer disease, or diabetes (14 -16). Furthermore, TKT was suggested to be a critical determinant for impaired hippocampal neurogenesis, lymphatic metastasis of hepatocarcinoma, fibromyalgia, tumor cell growth, resistance of colon cancer toward 5-fluoruracil, and female fertility (17)(18)(19)(20)(21).…”
