The thiopurine antimetabolites 6-thioguanine and 6-mercaptopurine are important chemotherapeutic drugs in the treatment of childhood acute lymphoblastic leukaemia. Measurement of metabolites of these thiopurines is important because correlations exist between levels of these metabolites and the prognosis in childhood acute lymphoblastic leukaemia. The reversed-phase method for the determination of extracellular thiopurine nucleosides and bases was previously developed and has been modified such that methylthiopurine nucleosides, bases, thioxanthine and thiouric acid can be measured also. The anion-exchange method enables the determination of intracellular mono-, di-and triphosphate (methyl)thiopurine nucleotides in one run. Extraction on ice with perchloric acid and dipotassium hydrogenphosphate results in good recoveries for (methyl)thiopurine nucleotides in lymphoblasts and peripheral mononuclear cells and for methylthioinosine nucleotides in red blood cells. Measure ment of the low concentrations of mono-, di-and triphosphate thioguanine nucleotides in red blood cells (detection limit 20 pmol/109 cells) is possible after extraction with methanol and methylene chloride, followed by oxidation of thioguanine nucleotides with permanganate and fluorimetric detection.

A sensitive and accurate reversed phase liquid chromatographic assay was developed for the determination of 6-mercaptopurine (6MP) (the active metabolite of azathioprine) in human plasma. The assay involved extraction into acetonitrile and dichloromethane from plasma pretreated with 0.038 M of dithiothreitol solution. The residue was analyzed by isocratic chromatography on a C18 analytical column with UV detection at 326 nm. The average extraction recovery of 6MP was 85%. The method has been applied successfully to the determination of 6MP and its metabolites in pharmacokinetic studies.

The electrochemical detection of 6-mercaputopurine (6-MP), which is one of the most well-known drugs of antimetabolites, and its analogues was studied. The presented method is based on chemisorption (accumulation)/ desorption processes of drugs onto/from the Au electrode surface. Using this method, we succeeded in detecting 6-MP even in very diluted solution (concentration of 6-MP: 1 Â 10 À8 M). The lower detection limit of 6-MP was considerably improved compared with the previous HPLC studies by using our proposed system. Desorptive potentials of 6-MPmetabolites and its analogues were different from those of 6-MP. This provides the possibility of simultaneous detection of 6-MP and its typical metabolites.
