High MMP-1, MMP-2, and MMP-9 protein levels in osteoarthritis

Abstract: ABSTRACT. Our study examined the relationship between the expression of matrix metalloproteinases (MMP)-1, MMP-2, and MMP-9 proteins and the pathogenesis of osteoarthritis (OA). We employed rigorous inclusion and exclusion criteria in computer-based bibliographic databases to extract published studies relevant to this investigation. The STATA 12.0 software was used for the statistical analyses. A total of 1408 studies were initially searched, and 10 studies with 458 OA patients and 295 healthy controls were in… Show more

“…These studies have demonstrated the enhanced production of key inflammatory mediators from chondrocytes exposed to OA synovial fluid or the synovium itself, which may act to further stimulate macrophage inflammation. 12,56,57,68,69 Cumulatively, our results agree well with changes seen in early OA and with similar in vitro model systems using NCs.…”

“…Each of these enzymes and inflammatory cytokines has been shown to be increased in OA. 5,12,56,57 These results demonstrate significant initial phenotypic differences between the NCs and OACs before coculture with macrophages.…”

“…MMP1 is induced in synovial fibroblasts in response to proinflammatory mediators such as IL1b and TNFa 21 . Synovium, as well as chondrocytes, might contribute to increased synovial fluid MMP1 levels observed in OA 22 . Association of MMP1 expression with symptomatic disease is unlikely to be entirely explained by cartilage structural damage because our cases and controls were matched for severity of macroscopic chondropathy.…”

“…MMP‐3 was found to aggravate inflammation by activating a variety of pro‐MMPs and cleaving extracellular components . MMP‐1 and MMP‐13 could break down type II collagen, proteoglycans, and other extracellular matrix components in cartilage . Furthermore, IL‐1β can induce the synthesis of MMPs in chondrocytes .…”

“…25 MMP-1 and MMP-13 could break down type II collagen, proteoglycans, and other extracellular matrix components in cartilage. 26,27 Furthermore, IL-1β can induce the synthesis of MMPs in chondrocytes. 28 In this study, we found that pretreatment with salidroside efficiently suppressed IL-1β-induced production of MMP-1, MMP-3, and MMP-13 in OA chondrocytes.…”
