High-frequency shoot regeneration through transverse thin cell layer culture in Dendrobium Candidum Wall Ex Lindl.

Abstract: An efficient in vitro propagation protocol for Dendrobium candidum Wall ex Lindl. using transverse thin cell layer (tTCL) culture system was established. The frequency of shoot regeneration and the number of adventitious buds produced from the regenerated shoots significantly relied on the concentration of plant growth regulators, and the position and orientation of the explant. Murashige and Skoog (MS) medium with half-strength macronutrients and 2% sucrose, supplemented with 1.2 mg l -1 naphthaleneacetic aci… Show more

“…produced five shoots per explant when treated with half-strength MS supplemented with 2 mg l -1 BA, 0.2 mg l -1 NAA, and 0.1 mg l -1 IBA (Luo et al 2006). The maximum number of adventitious buds per explants that has been reported to date-an average of 24.5-were generated from Dendrobium candidum explants treated with 1.2 mg l -1 BA and 1.2 mg l -1 NAA (Zhao et al 2007). Results from Martin (2003), Chen et al (2004a), Thomas and Michael (2007), and Hong et al (2008) show that, in most cases, Paphiopedilum spp.…”

“…Thus, mass propagation of this endangered species via somatic embryogenesis is an important approach for enlarging its population and for genetic reformation. For this purpose, we have established two efficient plant-regeneration systems for the mass propagation of D. candidum through somatic embryogenesis and organogenesis (Zhao et al 2007(Zhao et al , 2008. Based on the experimental system, it is possible to investigate molecular mechanisms involved in somatic embryogenesis in D. candidum.…”

“…Dendrobium candidum Wall ex Lindl. is one of the most popular orchids in South and Southeast Asia, and valued for its attractive flowers and medicinal uses (Zhao et al 2007;Shiau et al 2005). Dried stems are used in traditional Chinese or folk medicine as Yin tonic to nourish the stomach, promote secretion of body fluids, prevent the development of cataracts, relieve throat inflammation and fatigue, reduce peripheral vascular obstruction, and enhance immunity (Bao et al 2001).…”
