High frequency plant regeneration and accumulation of tropane alkaloids in regenerated plants of Scopolia parviflora

Abstract: An efficient in vitro plant regeneration system was established from callus culture of Scopolia parviflora. Callus was induced from adventitious roots on B5 medium with 0.45-9.04 μM 2,4-dichlorophenoxyacetic acid (2, 4-D). In vitro plantlet regeneration was achieved on B5 medium supplemented with 44.38 μM benzyladenine (BA), 3% sucrose, and 0.38% gelrite. Plantlets were transplanted to artificial soil and grown to maturity successfully in a greenhouse. The tropane alkaloid contents in regenerated plants were a… Show more

“…A BA concentration higher than 17.7 lM did not improve shoot proliferation, but reduced the percentage of bud break to a greater extent. Cytokinins are most critical for shoot induction and elongation in several systems including medicinal plants such as Clitoria ternatea (Rout 2005), Scopolia parviflora (Kim et al 2008) and Melothria maderaspatana (Baskaran et al 2009). Moreover, it is well known that BA promotes shoot formation in vitro.…”

“…Scopolamine content in de-differentiated tissues was high compared to that of hyoscyamine. As in an earlier study of Kim et al, the scoplolamine content in regenerated plants or organogenic calli was high compared to hyoscyamine. This result supposes that scopolamine biosynthesis is influenced by the cell differentiation process.…”

“…The adventitious roots of S. parviflora were induced from the rhizome of mature plants as reported . The organogenic and nonorganogenic calli were selected from adventitious root-derived calli …”

“…In plant tissue and cell culture analysis, hyoscyamine and scopolamine are measured by HPLC-DAD usually after separation on a RP18 column in an acetonitrile-buffer solvent system. Quantitation is mostly performed between 210 and 220 nm [72][73][74][75][76][77][78][79][80]. Control of pharmaceutical preparations containing atropine is possible by HPLC and DAD detection [81,82].…”
