High efficient chemiluminescent immunoassays for the detection of diclazuril in chicken muscle based on biotin–streptavidin system

Abstract: A simple, sensitive and specific chemiluminescence enzyme immunoassay based on biotin-streptavidin system (BASA-CLEIA) for diclazuril was established, which was about 2 and 4 times more sensitive than the traditional chemiluminescence enzyme immunoassay (CLEIA) and enzyme-linked immunosorbent assay. In the optimized generic assay, the half maximal inhibitory concentration (IC 50) of the BASA-CLEIA for diclazuril was 0.48 μg/kg. The limit of detection for diclazuril was 0.02 μg/kg. This method showed no cross r… Show more

“…To meet the needs of on-the-spot, rapid, low-cost, and accurate detection, immunological detection methods have become the main research focus. The existing reports of immunological detection methods include gold nanoparticle-based strips ( 25 ), gold nanoparticle-based lateral-flow strips ( 29 , 30 ), enzyme-linked immunoassays ( 24 ), and high efficiency chemiluminescent immunoassays ( 15 ). The accuracy and stability of immunological methods mainly depend on the antibody sensitivity.…”

“…2002-235 from the Ministry of Agriculture, China. In recent years, methods such as high-performance liquid chromatography ( 15 , 16 ), gas chromatography, and liquid chromatography tandem mass spectrometry ( 17 19 ) have been widely used to detect diclazuril residues. However, these methods are unsuitable for on-site spot, rapid, and batch inspections by enterprises and governments.…”

“…Chemiluminescence immunoassay (CLIA) is an assay that combines the chemiluminescence technique with immunochemical reactions and is widely used in clinical diagnosis because of its high sensitivity, rapidity, cost-effective, and easy automatization. ( Ishibashi et al, 2019 ; Li et al, 2020 ; Qi et al, 2020 ). However, a conventional fully automated CLIA analyzer is routinely performed in professional clinical laboratories equipped with huge instruments and paramedics ( Cetin et al, 2014 ; Yan et al, 2019 ), which limits the application of CLIA in large-scale screening of cancer in resource-limited settings, such as the MAGLUMI ® 800 CLIA analyzer ( Luang et al, 2020 ) and the Abbott i2000 CLIA analyzer ( Kim et al, 2007 ).…”
