Hetacillin: A Chemical and Biological Comparison with Ampicillin

Abstract: Hetacillin breaks down in aqueous solution to give ampicillin and acetone, both of which products have been identified chemically. The rate of breakdown is maximal between pH values 5 and 8, and is considerably slower under more acid conditions. The finding that hetacillin does not react with hydroxylamine at pH 7, taken in conjunction with other data presented, suggests that the antimicrobial activity of hetacillin may be low or even neglibible, and that it must be converted to ampicillin to show significant … Show more

“…Hetacillin is a β-lactam antibiotic that was used to treat bacterial infections (Hardcastle Jr et al 1966 ; Smith and Hamilton-Miller 1970 ). It was later withdrawn as it offered a lesser therapeutic value than the ampicillin derivatives.…”

“…Concentrations of 10 and 1 ,g of hetacillin/ml with j-lactamase did not inhibit, whereas similar concentrations of ampicillin did. (2,5,6) because they used methods which were too slow to show inhibition of growth occurring within the hydrolysis time of the antibiotic.…”

“…It is unstable under conditions of acidity and temperature suitable for antibiotic action, breaking down to ampicillin and acetone (2,5,6). The half-life of hetacillin at 37 C at pH 7.0 has been estimated at 15 to 30 min (2,6).…”

“…Ampicillin, a semi-synthetic: antibiotic active against many Gram-positive and Gramnegative bacteria, is widely prescribed in veterinary medicine for treating pneumonia and septicaemia. In relation to plasma and serum concentrations the pharmacokinetics of ampicillin and of its derivatives are well understood (Smith & Hamilton-Miller, 1970;Tan et al, 1974;Ziv et al, 1977). Ampicillin is also given orally in neonatal diarrhoea therapy in the calf (Jones, 1974;Rolinson & Stevens, 1961).…”
