Objective: This study was carried out to evaluate the antioxidant activity and effect of a hot water extract of Lycium chinense fruit (LFE) on liver regeneration induced by Partial Hepatectomy (PH) in rats. Methods: Antioxidant activity of LFE was determined. Rats were divided into three groups; laparotomized (sham, n=15), 70% PH (control, n=25), and LFE-treated after PH (experimental, n=35). Starting 7 days before PH, rats received DW or LFE (500 mg/kg in DW) orally once a day until sacrifice. Rats were sacrificed on d 0.5, 1, 2, 3, and 7 after surgery. Liver regeneration ratio and Proliferating Cell Nuclear Antigen (PCNA) expression were determined through immunohistochemistry and western blotting. Results: Final extraction yield of LFE was 36.0%. LFE scavenged the DPPH and the ABTS radicals in a concentration-dependent manner. The IC 50 of LFE on DPPH and ABTS antioxidant activity was 5.97 mg/ml and 2.53 mg/ml, respectively. The extent of liver regeneration seen in the experimental group (91.29%) at 7 d after PH was greater than that in the control group (89.43%). After PH, the liver reconstruction process involving collapse, re-arrangement and re-modelling of hepatic tissue was observed to happen sooner in the experimental group. The number of PCNA-positive hepatocytes obviously increased in the experimental group. PCNA-positive hepatocyte ratio peaked at 2 and 3 d in the experimental and control groups, respectively. Conclusions: Administration of LFE had beneficial effects on the regenerative capacity of remnant liver tissue after 70% PH in rats, probably owing to its antioxidant and proliferative properties.

BACKGROUND:The aim of this study is to investigate the effects of Lycium chinense Miller fruit and its representative constituent betaine on reduction of immobility time and blood parameters in balb/c mice. METHODS AND RESULTS:We investigated the immobility time and the changes in aspects of blood biochemical parameters by the administration of L. chinense Miller fruit and its representative constituent betaine, after the forced swimming test. The immobility time was significantly reduced about 41.3% and 53.6%, respectively, in the animal of L. chinense Miller fruit and its representative constituent betaine-administrated group for 7 days, in comparison with that of the control group. The level of blood urea nitrogen was significantly decreased in L. chinense Miller fruit and its representative constituent betaine-treated group compared with the control group (P < 0.05), respectively. In addition, the interlukin-2 levels of mice in L. chinense Miller fruit and betaine treated group was increased compared with the control group. Carbon tetrachloride (CCl4)-induced liver injury causes necrosis and fibrosis, which may lead to cirrhosis and liver failure. Although a lot of study to understand the mechanism of liver fibrosis, our knowledge of the molecular level of this disease is still incomplete. The aim of this study was to investigate the effect and regulation of nitric oxide (NO) and glial fibrillary acidic protein (GFAP) on the severity of hepatic injury in male Wistar albino rats with CCl4-induced hepatotoxicity and elucidate the underlying mechanism. A total of 20 male rats were randomly divided into healthy control group (A) and CCl4-induced hepatic fibrosis model group (B), with 10 rats in each group. The results of this study suggest CCl4-induced hepatic injury is mediated by excessive NO production and up-regulated by inducible and endothelial NO synthase (P<0.05), which may plays a role in increasing hepatic injury. Additionally, liver injury induced by CCl4 demonstrated significant increases in GFAP (P<0.05). Histological and immunohistochemical analyses of the CCl4-treated group exhibited increased inflammatory process and liver necrosis/fibrosis. Inflammatory response especially NO and GFAP expression is identified to play an important role in the development of liver injury and fibrosis induced by CCl4. Günümüzde karaciğer fibrozis mekanizmasının anlaşılmasına yönelik çok fazla çalışma olmasına rağmen, oluşum mekanizması ile ilgili moleküler düzeydeki bilgiler hala eksiktir. Bu çalışmada CCl4 ile hepatotoksisite oluşturulan erkek Wistar albino sıçanlarda fibrozisin oluşum aşamasında nitric oksit (NO) ve glial fibriller asidik protein (GFAP) etkilerinin araştırılması ve altında yatan mekanizmanın aydınlatılması amaçlanmıştır. Çalışmada 20 adet erkek sıçan, her grupta toplam 10 sıçan olmak üzere sağlıklı kontrol grubu (A) ve CCl4 ile oluşturulan karaciğer fibrozu grubu (B) olarak rastgele ikiye ayrıldı. Histolojik ve immunohistokimyasal analizlerle CCl4 grubunda yangısal değişiklikler ile karaciğer nekroz/fibrozis tespit edildi. On iki hafta süreyle CCl4 verilerek oluşturulan karaciğer hasarının oluşumunda, indüklenebilen ve endotelyal NO sentazın (P<0.05) yüksek sunumlarına bağlı olarak artan NO üretiminin önemli bir rol oynadığı belirlendi. Ayrıca, CCl4 grubunda karaciğerde GFAP (P<0.05) sunumlarında önemli artış da gösterildi. Sonuç olarak, CCl4 kaynaklı karaciğer hasarı ve fibrozisin gelişiminde NO ve GFAP'ın önemli bir rol oynadığı tespit edildi.
