Hematologic Evaluation of Normal and Anemic Lambs with the Technicon H*1 Using EDTA or Heparin as Anticoagulants

Abstract: A study was performed to evaluate blood from young lambs using the Technicon H*1 hematology analyzer, with emphasis on RBC parameters, comparison of tripotassium EDTA and heparin, and the effects of storage on heparinized blood. Blood samples from lambs 2 days to 18 weeks of age were analyzed within 6 hours, revealing a high precision, except for WBC counts in heparinized blood. The HCT values estimated by the H*1 correlated well (r(2)=.90) with those obtained by the microhemotocrit method. Mean hematologic va… Show more

“…Experienced avian veterinarians working with experienced restrainers, who minimize trauma to the vessel wall, can collect a high quality sample without the addition of heparin. Reducing the amount of heparin in the sample is desirable when using automated hematology analyzers, such as the Cell Dyn 3500 (Abbott Laboratories, Abbott Park, Ill) because of a decreased coefficient of variation of the WBC count 1 as well as decreasing potential dilution and assay artifact in a biochemical sample. If most of the heparin is expelled from the needle hub, it will minimally affect the sample.…”

“…Haematocrit is increased in birth, declines up to the 14th day of life, partially because of iron deficiency (Ullrey et al 1965a, Schalm 1986, and after day 14 increases continuously Στην παρούσα μελέτη καταβλήθηκαν προσπάθειες ώστε να αποφευχθεί η επίδραση παραγόντων, όπως το αντιπηκτικό, η αποθήκευση των δειγμάτων, οι βελόνες και οι χειρισμοί της αιμοληψίας, στα αποτελέσματα των αιματολογικών εξετάσεων.

“…EDTA is preferred for the complete blood count compared to heparin and sodium citrate (Vatn et al 2000, Morris 2002a). Vacuum glass tubes were filled with the proper amount of blood, in order the correct analogy of blood and anticoagulant to exist.…”
