Heat-Shock Protein 70 as a Tumor Antigen for in vitro Dendritic Cell Pulsing in Renal Cell Carcinoma Cases

Abstract: Immunological functions of heat shock proteins (HSPs) have long been recognized. In this study we aimed to efficiently purify HSP70 from renal cell carcinoma and test it as a tumor antigen for pulsing dendritic cells in vitro. HSP70 was purified from renal cell carcinoma specimens by serial column chromatography on Con A-sepharose, PD-10, ADP-agarose and DEAE-cellulose, and finally subjected to fast protein liquid chromatography (FPLC). Dendritic cells derived from the adherent fraction of peripheral blood mon… Show more

“…Similarly, in patients with colorectal cancer treated by local hyperthermia, exosomes derived from tumor cells are able to stimulate dendritic cells to secrete more IL-6 promoting the differentiation of Th 17 regulatory T-lymphocytes; resulting in an anti-tumor effect [63]. HSP 70 secreted by the cancer cell after chemical [64] or heat exposure [65] acts like a DC antigen enhancing immunity against the tumor through Toll Like Receptor (TLR) 4. HSP 90 also accelerates DC maturation (see Figure 2) [66].…”

“…Previous studies also suggested the extraordinary role of Hsp70 in DC activation. Hsp70 is secreted by adherent tumor cells upon heat shock reaction and acts as a DC antigen, which increases immunity against tumor cells through TLR4 in DCs [ 59 , 60 ]. This role of Hsp70 was reported in glioblastoma, hepatocarcinoma, and melanoma cells, and in some clinical studies [ 61 , 62 , 63 , 64 , 65 ].…”
