the current study aimed to formulate Selenium-chitosan-Mupirocin (M-Senps-ccH) complex. the nanohybrid system was prepared using chitosan-cetyltrimethylammonium bromide (ctAB)based hydrogel (ccH) that entrapped mupirocin (M) and selenium nanoparticles (Senps). the in vitro studies were performed by evaluation of the antibacterial activity and toxicity on L929 mouse fibroblast cell line. The in vivo study was conducted on rat diabetic wound infection model that was infected by mupirocin-methicillin-resistant Staphylococcus aureus (MMRSA). the wounds were treated by M-SeNPs-CCH nanohybrid system with concentrations of M; 20 mg/ml, CCH; 2 mg/ml and SeNPs; 512 μg/ml in two times/day for 21 days. The therapeutic effect of this nanohybrid system was evaluated by monitoring wound contraction and histopathological changes. evaluation of the average wound healing time showed a significant difference between the treatment and control groups (P≤0.05). The histopathological study indicated that the amount of wound healing was considerable in M-Senps-ccH nanohybrid system groups compared to the control and M groups. the M-Senps-CCH nanohybrid system formulated in this study was able to reduce 3-fold MIC of mupirocin with synergistic antibacterial activity as well as to play a significant role in wound contraction, angiogenesis, fibroblastosis, collagenesis, proliferation of hair follicle, and epidermis growth compared to the control group (P ≤ 0.05). This research suggests that this nanohybrid system might be a development for the treatment of diabetic wound infection at mild stage. Management of the infectious diabetic foot ulcer remains a global health issue 1,2. According to the World Health Organization (WHO), about 422 million people worldwide suffer from diabetes 2. About 25% of diabetic patients experience diabetic foot wounds in their lifetime 3,4 , half of them suffer infection 3. Diabetic wounds are classified into three groups, i.e. mild, moderate and severe 3. To avoid amputation and reduce health care costs, it is essential to control the infection 5. Gram-positive bacteria such as Staphylococcus aureus and beta-hemolytic streptococci are common infectious agents in the mild stage of diabetic foot wounds 3,5. Background: Cancer is currently the second leading cause of death worldwide that is originated from cell growth and proliferation without control. Acute lymphoblastic leukemia (ALL) is one of the types of leukemia that affects lymphocyte maturation and it is common among children. Silver nanoparticles are considered one of the targeted chemotherapy methods by creating cytotoxicity. Objectives: In this research, a comparative study of cytotoxic effect of silver nanoparticles was evaluated on human lymphocytes and HPB-ALL cell line as an in vitro study. Methods: In this experimental study, lymphocytes and HPB-ALL cell line were exposed to silver nanoparticles at RPMI 1640 medium culture in order to assess toxicity for 24 hours. To this aim, MTT assay was used to evaluate the toxicity of the silver nanoparticles. DNA fragmentation and apoptosis were evaluated by Gel Electrophoresis and Flow Cytometry, respectively. Moreover, quantitative PCR was performed on bax, bcl-2, and caspase-9 genes. Results:The results of MTT assay showed IC50 values of silver nanoparticles were 5.87 and 2.68 µg/mL for lymphocytes and HPB-ALL cell line, respectively. The results showed that silver nanoparticles could split DNA of the HPB-ALL cell line more than DNA of the lymphocytes during DNA fragmentation. Flow Cytometry results indicated that the early apoptosis was 6.04% and 22.75% in lymphocytes and HPB-ALL cell line, respectively. Moreover, Q-PCR results showed a significant up-regulation of caspase-9 and bax genes and downregulation of bcl-2 gene in comparison to the control group. Conclusions: The silver nanoparticles had cytotoxic effects on the lymphocytes and HPB-ALL cell line. The results showed that the silver nanoparticle had a significant cytotoxic effect on HPB-ALL cell line.

Prospects for the use of a drug based on a complex of fatty acid esters in models of laboratory animals in vivo

In this work we carried out studies of the first obtained micellar composition of fatty acid esters with allantoin on a water basis to determine its local irritant effect and therapeutic efficacy in the treatment of burns. It was found that there was a sequential change in the phases of the burn wound process in all groups. During the observation period, wound suppuration was not observed; there were 4 cases of a complicated course of the wound process and a fatal outcome in the control group. When conducting studies of local irritant effects, the assessment was carried out in points, in the experimental group it was on average 2 points as for the animals of the control group. The conducted studies allow us to conclude that the drug, when applied to the skin according to the interstate standard GOST ISO 10993-10—2011, has a weak degree of response to irritation in guinea pigs. The application of a permissive dose of the drug to sensitized guinea pigs does not cause a response from the skin, which indicates the absence of allergenic properties in this composition.
