Synergistic Antibacterial Activity and Wound Healing Properties of Selenium-Chitosan-Mupirocin Nanohybrid System: An in Vivo Study on Rat Diabetic Staphylococcus aureus Wound Infection Model

Abstract: the current study aimed to formulate Selenium-chitosan-Mupirocin (M-Senps-ccH) complex. the nanohybrid system was prepared using chitosan-cetyltrimethylammonium bromide (ctAB)based hydrogel (ccH) that entrapped mupirocin (M) and selenium nanoparticles (Senps). the in vitro studies were performed by evaluation of the antibacterial activity and toxicity on L929 mouse fibroblast cell line. The in vivo study was conducted on rat diabetic wound infection model that was infected by mupirocin-methicillin-resistant St… Show more

“…An increase of tissue force with higher resistance of the scar due to organization of collagen may justify this detected delay ( 68 ). Without evident detrimental effects on healing ( 69 ), MU could injure keratinocytes and fibroblasts of healthy skin ( 68 ) what stimulates new formulations ( 70 ) or synergistic antimicrobial combinations with best healing properties ( 71 ). Hyperkeratosis/clinical desquamation in MU-wounds shows clinical-histological correspondence with an excess of keratin production and superficial cutaneous desquamation in nude mice ( 23 ); this could have a double explanation: uncontrolled wound healing in untreated wounds or changes of lipid layer of skin as secondary effect of repetitive dose of topical MU in humans (drug commercial package) and possibly in SKH-1 mice.…”

“…The therapeutic combination within the hydrogel demonstrated synergic activities against mupirocin‐methicillin‐resistant S. aureus (MMRSA) inoculated in an injury created in a diabetic rat model, while improving the wound healing associated with the bacterial infection, stimulating epidermal growth, and decreasing the associated inflammation, suggesting its potential for treatment of diabetic wound infections. [ 204 ] SeNPs produced by biosynthesis using Lactococcus lactis had a protective effect on intestinal infection by E. coli , preserving the intestinal epithelial barrier due to its antioxidant and anti‐inflammatory effects and by increasing occludin an claudin‐1 expression. “…The incorporation of antibiotics into chitosan hydrogels can enhance their efficacy in treating wound infections. For example, mupirocin was added to chitosan-cetyltrimethyl ammonium bromide hydrogels to effectively resist infections caused by S. aureus and Streptococcus pyogenes ( S. pyogenes ) ( Golmohammadi et al, 2020 ).…”
