Hapten Synthesis for the Development of a Competitive Inhibition Enzyme-Immunoassay for Thiram

Abstract: An enzyme-linked immunosorbent assay (ELISA) was developed for the fungicide thiram. Two types of haptens were synthesized. The first type exhibits the two symmetrical N-alkyl dithiocarbamate patterns of thiram with a spacer arm linked to one of the N-methyl terminal group. The second type exhibits one of the two symmetrical N-alkyl dithiocarbamate patterns of thiram with a variable-length spacer arm linked to one sulfur atom. Polyclonal antibodies suitable for thiram detection were obtained from immunization … Show more

“…On the other hand, experience in antibody production against small aliphatic structures in hapten immunochemistry is limited. A sporadic attempt to produce antibodies against an underivatised small hapten structure was accomplished by Gueguen et al using immunogens with a di-thiocarbamate group attached to a carrier protein [22]. Surprisingly, antibody formation against small thiram pesticide was evoked, although antibodies exhibited very low sensitivity when incorporated into an indirect ELISA system.…”

“…Black polystyrene high-binding plates were from Costar (Cambridge, Massachusetts). The hapten-protein conjugate (OVA-2C) and the polyclonal antibodies against thiram were prepared as previously described (Gueguen 2000). 5…”

“…Various analytical methods to determine both qualitatively and quantitatively thiram have been reported, based on spectrophotometric (Tunceli 2001) chromatographic (Gustafsson on 1981;Brandsteterova 1986) and immuno-enzymatic (Gueguen 2000;Queffelec 2001;Aulakh 2005) techniques.…”

“…Many methods like spectrophotometry, 1,[3][4][5][6][7][8] flame atomic absorption spectrometry 9,10 FT-IR, 11 gas chromatography, 12,13 HPLC, [14][15][16] polarography, 17,18 biosensors, 19 and enzyme linked immuno sorbent assay 20 are available for Zineb determination and other dithiocarbamates. Most of the applied methods use different techniques like spectrophotometry and gas chromatography 3 based on analysis of CS 2 , H 2 S or amines which are evolved by the decomposition of EBDCs.…”
