Halofuginone, a promising drug for treatment of pulmonary hypertension

Abstract: Background and Purpose Halofuginone is a febrifugine derivative originally isolated from Chinese traditional herb Chang Shan that exhibits anti‐hypertrophic, anti‐fibrotic and anti‐proliferative effects. We sought to investigate whether halofuginone induced pulmonary vasodilation and attenuates chronic hypoxia‐induced pulmonary hypertension (HPH). Experimental Approach Patch‐clamp experiments were conducted to examine the activity of voltage‐dependent Ca2+ channels (VDCCs) in pulmonary artery smooth muscle cel… Show more

“…Anesthetized (by intraperitoneal injection of 120 mg/kg sodium pentobarbital) C57Bl/6 mice were used for whole lung angiogram experiments based on the published protocol (36). Heparin (20 IU) was injected, at first, into the RV to prevent blood clotting, then a polyethylene tube was inserted into the main pulmonary artery (PA) through the RV to perfuse PBS (pH = 7.4 at 36 C-38 C) to washout residual blood in the lung.…”

“…Chang Shan ( Dichroa febrifuga Lour ) is a traditional Chinese medicine as a treatment for malaria, dating back about 2000 years of history ( Zhou et al, 2013 ). Halofuginone is a natural quinazolinone alkaloid extracted from the plant Dichroa febrifuga ( Jain et al, 2021 ). Halofuginone is less toxic to normal cells, but it significantly inhibits the growth of CRC and induces reactive oxygen species (ROS) production and cell apoptosis in a dose-dependent manner ( Chen et al, 2015 ).…”

“…Halofuginone, a minor alkaloid extracted from the Chinese traditional herb Dichroa febrifuge , has significant anti-tumor, anti-hypertrophic, and anti-fibrotic properties ( Pines and Spector, 2015 ; Jain et al, 2021 ). Xia et al investigated the mechanism of Halofuginone inhibiting MCF-7 cell growth in vitro from the perspective of exosomes ( Xia et al, 2019 ).…”
