Transportation stress can result in significant economic losses to producers due to decreased animal productivity and increased medication costs associated with sickness such as bovine respiratory disease (BRD). Meloxicam (MEL) provides pain relief and anti-inflammatory effects in cattle for several days after a single oral treatment. Our hypothesis was that MEL administration before shipping would reduce the impact of long-distance transportation on circulating physiological biomarkers of stress and inflammation in beef steers. Ninety-seven beef steers were blood sampled for baseline biomarker determination and then randomly assigned to receive either 1 mg/kg MEL (n = 49) or a placebo (CONT; n = 48) per os before a 1,316-km transportation event lasting approximately 16 h. Calves were then blood sampled on arrival and 5 d later. Changes in the hemogram, circulating plasma proteins, total carbon dioxide (TCO 2 ), fibrinogen, substance P (SP), cortisol, haptoglobin (Hp)-matrix metalloproteinase-9 (MMP-9) complexes, and tumor necrosis factor α (TNFα) between treatments over time were compared using a mixed effects model with statistical significance designated as P < 0.05. Analysis of covariance was conducted to assess the relationship between circulating MEL concentrations and biomarker changes over time. An increase in neutrophil, platelet, monocyte, white blood cell, and red blood cell counts occurred after transportation (P < 0.0001) and a decrease in lymphocyte count were observed (P < 0.0001). Meloxicam treatment reduced the stress-induced neutrophilia (P = 0.0072) and circulating monocyte count (P = 0.013) on arrival. Mean corpuscle hemoglobin (P = 0.05), mean corpuscle volume (P = 0.05), and lymphocyte count (P = 0.05) were also greater in the CONT calves compared with MEL calves after transportation. Furthermore, Hp-MMP-9 complexes, TCO 2 , TNFα, plasma proteins, and SP increased and cortisol decreased after shipping (P < 0.01). Meloxicam treatment tended to reduce serum cortisol concentrations (P = 0.08) and there was evidence of a time × treatment interaction (P = 0.04). An inverse relationship between plasma MEL concentrations and circulation cortisol concentrations (P = 0.002) and neutrophil (P = 0.04) and basophil counts (P = 0.03) was also observed. The results suggest that MEL administration may reduce the impact of long-distance transportation on circulating physiological biomarkers of stress and inflammation in beef calves. RESUMOO hemograma e o perfil bioquímico são usados para diagnosticar doenças em animais domésticos. Esses exames podem ser influenciados pela idade, sexo, nutrição, raça, espécie e condições ambientais. Portanto, dados de uma região não podem ser totalmente extrapolados para animais criados em regiões geograficamente distintas. Isso se aplica a ovinos criados no Bioma Amazônico. O objetivo deste estudo foi determinar os valores hematológicos e bioquímicos de ovinos Santa Inês, de diferentes idades e gêneros criados na Amazônia oriental. Foram examinados 91 ovinos divididos em três grupos: G1 (3-6 meses de idade, n = 31); G2 (7 a 24 meses de idade, n = 30) e G3 (mais de 24 meses de idade, n = 30). O hemograma e as determinações bioquímicas foram realizados com um contador automático e um analisador semi-automático, respectivamente. Os resultados foram comparados pelo teste de Tukey. O número de eritrócitos, índices eritrocitários, plaquetograma, número de eosinófilos, teor de proteína total, de ureia e de creatinina foram influenciados pela idade dos animais. O coeficiente de variação dos eritrócitos e a concentração de creatinina foram influenciados pelo sexo, sendo maiores nos machos. A relação neutrófilos:linfócitos (N:L) foi maior que um para todos os grupos etários. Neste estudo foram determinados valores de referência para ovinos criados na Amazônia Oriental. Além disso, demonstrou-se que ao interpretar exames hematológicos e bioquímicos de ovinos, a idade e o sexo devem ser considerados. Background: Shearing is considered as a stressful factor in sheep, capable of causing some behavioural changes including variations of body condition and hormonal and haematochemical parameters. On these basis, circulating ACTH and cortisol concentrations were determined in sheep before and after exposure to aversive stimuli in order to evaluate the potential use of these hormones to objectively measure physical and mental stress responses to isolation, restraint and shearing.Materials, Methods & Results: Forty Pinzirita sheep, used as experimental group, were evaluated in baseline conditions, 24 h before isolation, restraint and shearing (T0), after isolation (T1), after restraint and shearing (T2), by taking into account the influence of previous shearing experience (just shorn). A control group of forty sheep (intact) was evaluated in baseline condition (T0). According to previous experience the experimental subjects were divided in two groups, respectively group A represented by 20 sheep (10 males and 10 females), aged 1-2 years, without previous shearing experience (intact); and group B represented by 20 sheep (10 males and 10 females), aged older than 2 years, previously exposed to shearing (just shorn). A control group C of 40 sheep splitted into two sub-groups represented by 10 intact males and 10 intact females, aged 1-2 years, and 10 males and 10 females just shorn, aged older than 2 years, was evaluated in baseline conditions (T0). Blood samples were collected 24 h before experimental practices (T0: 08:00 h), after 1 h after of isolation (T1) and after restraint and shearing, around an average of 5 min (T2). All samples were taken in quiet conditions by the same operator. Serum ACTH concentrations were assayed by a commercial radioimmunoassay (RIA). Serum cortisol concentrations were assayed by amplified Enzyme Immunoassay (EIA). Two-way RM ANOVA showed a significant effect of husbandry procedures on ACTH changes, with higher ACTH and cortisol concentrations of just shorn than intact males (F = 15.76; P < 0.0001) and females (F = 17.34; P < 0.0005), and on cortisol changes, with higher cortisol concentrations of just shorn than intact males (F = 6.54; P < 0.01) and females (F = 22.66; P < 0.0001).Discussion: The highest ACTH and cortisol values after isolation, with an appreciable increase especially after restraint and shearing in just shorn sheep, showed that the previous shearing exposure could not induce a habituation to the stimulus, probably promoting a larger release of ACTH and cortisol hormones, so inducing an early and marked coping response to stress in just shorn specimens. Thus, it would appear that different ACTH and cortisol responses of sheep with or without previous shearing experience over the study period resulted in some significant differences when exposed to isolation, restraint and shearing, probably due to changes in pituitary-adrenal axis reactivity. Nevertheless, it is possible to interpret the increase in ACTH and cortisol concentrations related to intrinsic endocrine control mechanisms as an increase due to a rise in the stressful stimulus or coping response. The magnitude of ACTH and cortisol changes following isolation, restraint and shearing suggests that measurements of both hormones are useful in assessing husbandry procedures stress in sheep, with marked elevations in just shorn sheep, indicating that experience can induce an early much larger coping response to stressful practice of shearing.
