Gold nanocluster-based ratiometric fluorescent probes for hydrogen peroxide and enzymatic sensing of uric acid

Abstract: A method is described for ratiometric fluorometric assays of HO by using two probes that have distinct response profiles. Under the catalytic action of ferrous ion, the 615 nm emission of protein-stabilized gold nanoclusters (under 365 nm photoexcitation) is quenched by HO, while an increased signal is generated with a peak at 450 nm by oxidizing coumarin with the HO/Fe(II) system to form a blue emitting fluorophore. These decrease/increase responses give a ratiometric signal. The ratio of the fluorescences at… Show more

“…H 2 O 2 has been reported to be a weak fluorescence quencher of AuNCs in previous literature. 55 The Fmoc-FF/AuNCs/HRP films showed a luminescence decrease phenomenon after dipping into pH 5.0 buffers containing H 2 O 2 (Figure 5A). The FL 660 from the films gradually declined as the H 2 O 2 concentration (c H 2 O 2 ) was increased from 0 mM to 2.0 mM and reached a plateau at 2.0 mM.…”

“…Apart from fluorescence quenching and enhancement, another strategy for biosensing is based on ratiometric fluorescence (RF)/color change [ 145 , 146 , 147 ]. In this strategy ( Figure 8 c), the protein-templated metal NC is typically used in conjunction with a secondary probe emitting at a different wavelength and, thus, a different emission color [ 133 , 148 , 149 ]. The analyte interacts with the protein-templated metal NC to quench its emission, leaving only the emission of the second emitter.…”

“…The fluorescence changes of F 450 / F 640 had fantastic linearity to H 2 O 2 concentrations in the range of 0.8−200 µmol L −1 ( R 2 = 0.9993), with LOD of 0.2 µmol L −1 . In another study, Au NCs‐based ratiometric fluorescent probes were synthesized for probing H 2 O 2 and proved to be dual emissive by Yan group . In the presence of ferrous ion as catalytic agent, the emission of protein‐stabilized Au NCs at 615 nm was quenched by H 2 O 2 , whereas the blue emission at 450 nm was enhanced due to oxidizing coumarin with the H 2 O 2 /Fe(II) system.…”
