Glutathione Metabolism, Mitochondria Activity, and Nitrosative Stress in Patients Treated for Mandible Fractures

Abstract: The aim of the study was to evaluate the effect of titanium bone fixations on mitochondrial activity, reactive oxygen species (ROS) production, glutathione metabolism, and selected markers of oxidative/nitrosative stress in the periosteum-like tissue of patients treated with mandible fractures. The study group consisted of 30 patients with bilateral fractures of the mandible body eligible for surgical treatment. Our study is the first one that indicates disturbances of mitochondrial activity as well as a highe… Show more

“…The contents of dityrosine, kynurenine, N-formylkynurenine and tryptophan were measured based on the characteristic fluorescence at 330/415, 365/480, 325/434 and 95/340 nm, respectively [ 38 ]. The results were normalized to the fluorescence of 0.1 mg/mL quinine sulphate in 0.1 M H 2 SO 4 [ 39 ].…”

“…The available evidence indicates that thiol groups of glutathione are in balance with thiol groups of proteins, which enables the maintenance of functional activity of proteins as well as a reduced status of protein thiol groups [47]. With 40% decrease in glutathione concentration in NWS and 16.7% in SWS, a positive correlation between glutathione concentration and salivary amylase activity in both salivary glands is therefore not surprising and neither is 58% decrease in activity of this enzyme in NWS and 36.7% in SWS.…”

“…Nitric oxide (NO) concentration was measured colorimetrically using sulfanilamide and NEDA•2 HCl (N-(1-naphthyl)-ethylenediamine dihydrochloride). Nitrate was converted to nitrite using nitrate reductase and total NO was measured [40,41]. The absorbance was measured at 490 nm and NO concentration was expressed in µmol/mg protein.…”
