Glucose Challenge Stimulates Reactive Oxygen Species (ROS) Generation by Leucocytes

Abstract: Diabetes mellitus is associated with increased ROS generation, oxidative injury and obesity. To elucidate the relationship between nutrition and ROS generation, we have investigated the effect of glucose challenge on ROS generation by leucocytes, p47phox protein, a key protein in the enzyme NADPH oxidase and alpha-tocopherol levels. Blood samples were drawn from 14 normal subjects prior to, at 1, 2 and 3 h following ingestion of 75 g glucose. ROS generation by polymorphonuclear leucocytes (PMNL) and mononuclea… Show more

“…This strong relation between blood glucose level and CAD made clinicians and researchers associate hyperglycemia and insulin resistance states to circulatory cell dysfunction. A study done by Mohanty et al revealed an increase in ROS by leukocytes after 75grams glucose challenge [142]. This was also confirmed by Dandona et al after giving 30 grams of glucose in his study [143].…”

“…We evaluated TOS and TAS levels at 5th hour after performing OGTT and MMT. Given that ROS generations were demonstrated to increase at around 1st and 2nd hours, and progressively to decrease in other studies [22,26], our drawing blood samples at 5th hour to evaluate TAS and TOS levels may be a mistaken effort.…”

“…Ceriello et al showed for the first time that TAS levels decrease after OGTT and MMT in normal individuals and patients with type 2 DM [40,41]. Additionally, Mohanty et al [26]. reported that after loading 14 patients 75 gr of glucose, the production of reactive oxygen species (ROS) increases p47 phox (an integral protein component of NADPH oxidase) and thiobarbituric acid reactive substances (TBARS), a sign of lipid peroxidation while decreasing the level of alpha tocopherol.…”

“…Interestingly, macrophages from diabetic mice display a proinflammatory phenotype when compared with non-diabetic mice [40]. In addition, glucose challenge in human subjects increases ROS production and p47 phox expression in leucocytes [41]. Accordingly, analyses of the draining lymph nodes of the aortic sinus revealed increased T-cell activation in the setting of diabetes.…”
