Glucocorticoids inhibit human hematopoietic stem cell differentiation toward a common ILC precursor

Abstract: Background: Innate lymphoid cells (ILCs) comprise cytotoxic natural killer (NK) cells and helper ILCs (hILCs). Human hILC development is less characterized as compared with that of NK cells, although all ILCs are developmentally related. It has been reported that the immunosuppressive drugs glucocorticoids (GCs) regulate ILC function, but whether they control ILC differentiation from hematopoietic stem cells (HSCs) is unknown. Objectives: This study sought to analyze the effect of GCs on ILC development from HS… Show more

“…206 In line with this finding is GC-induced upregulation of PRDM1, which encodes BLIMP-1, a gene that promotes differentiation of B cells to plasma cells. 206 GCs also favor the generation of NK cells from myeloid precursors, 221 whereas their proliferation, as well as their ability to lyse tubular endothelial cells, was significantly inhibited by prednisolone. 96…”

“…In addition to impacting bone biology, BMAds and GC have independent effects on hematopoiesis (39)(40)(41)(42). To determine if loss of GR in BMAds impacts hematopoiesis, we measured complete blood counts from male and female BMAd-Nr3c1 +/+ and BMAd- Nr3c1 -/mice.…”

“…In our example panel (Figure 2), we used CD3, CD19, and CD14 to exclude T cells, B cells and monocytes, respectively [16]. To make a more accurate exclusion of non‐ILCs, it is recommended to also include in the lineage CD34, TCRαβ, TCRγδ, CD123, CD15, and FcεRI.…”
