Germline transformation of the diamondback moth, Plutella xylostella L., using the piggyBac transposable element

Abstract: The diamondback moth, Plutella xylostella, is one of the most economically important agricultural pests. The larvae of this moth cause damage by feeding on the foliage of cruciferous vegetables such as cabbage, broccoli, cauliflower and rapeseed. Control generally comprises chemical treatment; however, the diamondback moth is renowned for rapid development of resistance to pesticides. Other methods, such as biological control, have not been able to provide adequate protection. Germline transformation of pest i… Show more

“…The total cost connected with the damage and the management of DBM was previously estimated at 4–5 billion USD per annum worldwide 2 . Much research has been devoted to the DBM, covering pest management strategies 3,4 , the mechanism of pesticide resistance 57 , its developmental biology 810 , and herbivory mechanism 11 . However, DBM outbreaks in Southeast Asia sometimes cause more than a 90% crop loss, and this pest still seriously threatens vegetable production in China.…”

“…The nonautonomous piggyBac vector allows germ-line transformation of insects via microinjection with separately supplied piggyBac transposase. The fluorescent marker is DsRed2 under the control of a baculovirus-derived promoter, Hr5ie1; this provides strong red fluorescence in a range of insect species (25,29,33,34). The conditional female-specific gene is designed to derive its lethal effect via hyperaccumulation of tTAV from a positive feedback loop, leading to transcriptional suppression (25,26).…”
