Gentamicin-Loaded Wound Dressing With Polyvinyl Alcohol/Dextran Hydrogel: Gel Characterization and In Vivo Healing Evaluation

Abstract: To develop a gentamicin-loaded wound dressing, cross-linked hydrogel films were prepared with polyvinyl alcohol (PVA) and dextran using the freezing-thawing method. Their gel properties such as gel fraction, swelling, water vapor transmission test, morphology, tensile strength, and thermal property were investigated. In vitro protein adsorption test, in vivo wound healing test, and histopathology were performed. Dextran decreased the gel fraction, maximum strength, and thermal stability of hydrogels. However, … Show more

“…Dextran is constituted by a(1?6)-linked D-glucose residues with some degree of branching via a(1?3) linkages, and is obtained from various bacterial strains via the action of dextransucrase [101,141]. Dextran is hydrophilic, highly soluble in water, inert in biological systems and easily functionalized through its reactive hydroxyl groups [195,196]. Dextran is available in a wide range of molecular weights, as well as in the form of several dextran derivatives.…”

“…It is biodegradable, biocompatible, resists protein adsorption and does not affect cell viability. Therefore it is a good polymer for medical applications such as bone, dermal and subcutaneous healing and for drug delivery [101,196]. It facilitates inflammatory cell infiltration and promotes angiogenic cell migration into the wounds [197].…”

“…It was recorded that these cross-linked hydrogels are capable of ideal water absorption and swelling and provide an ideal moist environment necessary for wound healing. In addition, it has been reported in the literature that cross-linking of polyvinyl alcohol with dextran and chitosan increases the flexibility and elasticity of the gel (Sung et al, 2010;Hwang et al, 2010). In another study, the novel thermoreversible wound gel formulation containing Polyhexadine showed good results for wound treatment and represented an alternative to silver sulfadiazine cream and iodine-based ointment, which are toxic and associated with many allergic reactions.…”
