Accumulation of misfolded proteins in the brain is a common hallmark of most age-related neurodegenerative diseases. Previous studies from our group identified the presence of anti-inflammatory and antioxidant compounds in leaves derived from the Chilean berry Ugni molinae (murtilla), in addition to show a potent anti-aggregation activity in models of Alzheimer´s disease. However, possible beneficial effects of berry extracts of murtilla was not investigated. Here we evaluated the efficacy of fruit extracts from different genotypes of Chilean-native U. molinae on reducing protein aggregation using cellular models of Huntington´s disease and assess the correlation with their chemical composition. Berry extraction was performed by exhaustive maceration with increasing-polarity solvents. An unbiased automatic microscopy platform was used for cytotoxicity and protein aggregation studies in HEK293 cells using polyglutamine-EGFP fusion proteins, followed by secondary validation using biochemical assays. Phenolic-rich extracts from murtilla berries of the 19–1 genotype (ETE 19–1) significantly reduced polyglutamine peptide aggregation levels, correlating with the modulation in the expression levels of autophagy-related proteins. Using LC-MS and molecular network analysis we correlated the presence of flavonoids, phenolic acids, and ellagitannins with the protective effects of ETE 19–1 effects on protein aggregation. Overall, our results indicate the presence of bioactive components in ethanolic extracts from U. molinae berries that reduce the load of protein aggregates in living cells.

It is predicted that 1.8 million new cancer cases will be diagnosed worldwide in 2020; of these, the incidence of lung, colon, breast, and prostate cancers will be 22%, 9%, 7%, and 5%, respectively according to the National Cancer Institute. As the global medical cost of cancer in 2020 will exceed about $150 billion, new approaches and novel alternative chemoprevention molecules are needed. Research indicates that the plants of the Lamiaceae family may offer such potential. The present study reviews selected species from the Lamiaceae and their active compounds that may have the potential to inhibit the growth of lung, breast, prostate, and colon cancer cells; it examines the effects of whole extracts, individual compounds, and essential oils, and it discusses their underlying molecular mechanisms of action. The studied members of the Lamiaceae are sources of crucial phytochemicals that may be important modulators of cancer-related molecular targets and can be used as effective factors to support anti-tumor treatment.

La industria alimentaria alrededor del mundo tiene la necesidad de sustituir los aditivos artificiales o sintéticos por alternativas naturales, debido a la demanda de los consumidores de ingredientes alimentarios naturales. En la búsqueda de proponer alternativas que coadyuven a esta problemática se evaluó la actividad hemolítica, antimicrobiana y antioxidante, así como la concentración de compuestos fenólicos totales y antocianinas monoméricas totales de extractos acuosos de cálices de jamaica de nueve variedades, cultivadas en los estados mexicanos Nayarit, Oaxaca y Puebla. La mayor concentración de compuestos fenólicos totales (CFT), antocianinas monoméricas totales (AMT) y actividad antioxidante por diversos métodos (DPPH·, ABTS·+, FRAP y ORAC) la presentó el extracto de la variedad de jamaica ‘cruza negra’ de Nayarit, la cual a una concentración de 20,000 µg/mL promovió una reducción en la velocidad de crecimiento de una cepa de Escherichia coli hasta en 51 % respecto al control. Por otra parte, la actividad hemolítica se mantuvo cercana al 1% aún con una concentración de 6000 µg de CFT/mL. Por lo anterior, se puede proponer el uso de los extractos acuosos de cálices de jamaica como un aditivo alimentario de origen natural, antioxidante, antimicrobiano y con muy baja actividad hemolítica.
