Genome wide microarray based expression profiles associated with Bm NPV resistance and susceptibility in Indian silkworm races of Bombyx mori

Abstract: The molecular mechanism involved in BmNPV resistance was investigated using a genome wide microarray in midgut tissue of Indian silkworm Bombyx mori. In resistant race (Sarupat), 735 genes up-regulated and 589 genes down-regulated at 12 h post BmNPV infection. Similarly, in case of susceptible race (CSR-2), 2183 genes up-regulated and 2115 genes down-regulated. Among these, nine up-regulated and eight down-regulated genes were validated using real-time qPCR analysis. In Sarupat, vacuolar protein sorting associ… Show more

“…Studies on this virus thus far, have demonstrated its proliferation within the host through a cascade of viral gene expression and identifying host resistant genes against BmNPV infection [2728]. However, the molecular mechanism of BmNPV-host interaction remains poorly understood.…”

“…In the insect fat body, Tret1 transports trehalose to the hemolymph. Although trehalose and Tret1 are thought to be produced primarily in the insect fat body, transcripts with similarity to Tret1 are frequently expressed by insect midgut epithelium [88,89]. Interestingly, trehalose, which is hydrolyzed by trehalase to produce two glucose molecules, is the first sugar utilized in the insect chitin biosynthesis pathway [35,90].…”

“…With the use of recently emerging data, researchers have begun to uncover the antiviral mechanisms in invertebrate immunity, particularly by screening for antiviral genes (Xia et al ., ; Bao et al ., ; Zhou et al ., ; Lekha et al ., ; Wang et al ., ; ; ; Xu et al ., ; Li et al ., ; Yu et al ., ). Whether these genes are resistance‐related genes remains unclear, however, and the exact antiviral mechanisms remain unknown.…”

“…We used Cuffquant and Cuffnorm components of cufflinks software to quantify the expression levels of transcripts and genes through the location information of mapped reads. We normalized Cuffquant and Cuffnorm via fragments per kilobase of transcript per million fragments mapped (FPKM) (Bao et al ., ; Lekha et al ., ; Wang et al ., ) using the formula FPKM = cDNA fragments/[mapped fragments (millions) × transcript length (kb)]. Genes with significant differences in the expression levels are referred to as DEGs or differentially expressed transcripts.…”
