Genome assembly and annotation of Periplaneta americana reveal a comprehensive cockroach allergen profile

Abstract: Background: One of the most common cockroach types in urban areas, the American cockroach (Periplaneta americana), has been reported to impose an increased risk of allergies and asthma. Limited groups of allergens (Per a 1-13) have been identified in this species due to the lack of genome-related information. Methods:To expand the allergen profile of P. americana, genomic, transcriptomic, and proteomic approaches were applied. With the support of a high-quality genome assembled using nanopore, Illumina, and Hi… Show more

“…Analysis of the brain transcritome SRAs from the American cockroach stung by the sphecid wasp Ampulex compressa (Arvidson et al, 2019) yielded one more sirp than those found in the first genome assembly of P. americana (Li et al, 2018), however this gene is present in two more recent genome assemblies from this species (Wang et al, 2022; Zheng et al, 2022).…”

“…The abundance of MA genes combined with the repeated sequences makes the assembly of transcripts and genes very difficult. The third Periplaneta genome assembly, obtained by a group working on cockroach allergens, still did not allow for their unambiguous assembly [ 31 ].…”

“…In brief, the protocol steps included: chopping the tissue into tiny pieces using a sterilized razor blade with a few drops of 40% glacial acetic then leaving it for 48 hours in the dark; immersing the processed slides into th fixation reagent (85:10:5 volumes of methanol:formaline:acetic acid); then hydrolysing them (using hydrochloric acid) and staining them (using Schiff’s reagent). We used three standards of known C-values: Periplaneta americana (3.41 pg) (61), Lasius niger (0.33 pg) (JF Flot, unpublished data), and Ambystoma mexicanum (32 pg) (62). A digital camera (5 megapixels) mounted on a compound microscope was used for imaging the slides, and nuclei measurements were performed using ImageJ (63).…”
