Brycon hilarii is a migratory fish widely distributed throughout the Paraguay River Basin. It is appreciated in sport fishing and for its superior meat quality. It is also the main species for tourist attraction in the Bonito region (State of Mato Grosso do Sul, Brazil). Considering the lack of information on the genetic structure of the fish of this species, the aim of the present study was to detect the genetic variability of Brycon hilarii through RAPD markers. A total of eighty specimens collected in different seasons at four sites of the Miranda River sub-basin (Paraguay River Basin, Brazil) were used for analysis. The results of genetic similarity, Shannon diversity, and AMOVA revealed differences between the sampling sites. Through AMOVA, differences between populations were more evident among the animals collected during the non-reproductive season, corresponding to a time of less movement of these fish. A population structuring model in which B. hilarii appears organized into genetically differentiated reproductive units that coexist and co-migrate through the studied system was suggested, contrasting the currently accepted idea that freshwater migratory fish form large panmictic populations in a determined hydrographic system. Despite the lack of a complete picture regarding the distribution of B. hilarii in the studied region, this initial idea on its population genetic structure could be an important contribution to providing aid for management and conservation programs of these fish.Keywords: Bryconinae, Characidae, conservation genetics, population genetics. Evidência genética de estruturação populacional do peixe neotropical de água doce Brycon hilarii ResumoBrycon hilarii é uma espécie de peixe migrador, distribuída por toda a Bacia do Paraguai, bastante apreciada tanto pela qualidade da carne quanto para a pesca esportiva, além de ser a principal espécie de interesse turístico da região de Bonito (MS, Brasil). Considerando a ausência de informações sobre a estrutura genética dos peixes desta espécie, o presente trabalho visou detectar a variabilidade genética de Brycon hilarii através de marcadores RAPD. Foi estudado um total de 80 exemplares amostrados em diferentes períodos do ano, em 4 localidades da sub-bacia do Rio Miranda (Bacia do Rio Paraguai, Brasil). Os resultados de similaridade genética, de diversidade de Shanon e AMOVA evidenciaram diferenças entre as localidades amostradas. Através da AMOVA, a diferenciação populacional foi mais evidente entre os peixes coletados no período não-reprodutivo, que representa a época de menor deslocamento desses peixes. Desta forma, ao contrário da idéia até então aceita de que os peixes migradores de água doce formam grandes populações panmíticas em um dado sistema hidrográfico, sugere-se a ocorrência de um modelo de sub-estruturação populacional onde os indivíduos de B. hilarii se organizam em unidades reprodutivas geneticamente diferenciadas e mantêm sua integridade co-existindo e co-migrando através do sistema estudado. Scaridae fish species including Scarus psittacus, S. frenatus, S. genazonatus, S. fuscopurpureus and S. niger, were collected from Red Sea in Egypt. The DNA fingerprints of these species were identified using RAPD-PCR technique with eight decamer primers. A total of 93 bands were scored in all species using the selected primers, with the size range varying from 190 to 2500 bp. On average, each primer amplified 11.63 bands per species. Each species had both monomorphic and polymorphic DNA bands. The polymorphic bands represent 34.41% in all fish species. The highest similarity index (70%) was found between Scarus psittacus and S. frenatus, while the lowest one (51%) was between Scarus psittacus and S. niger, S. genazonatus and S. fuscopurpureus and S. genazonatus and S. niger. Therefore the phylogenetic relationships among the five studied scaridae species were clearly resolved in dendrogram, they were clustered into four clusters groups: the first one comprised, S. fuscopurpureus and S. niger, the second included S. psittacus, S. frenatus and S. genazonatus, the third included S. genazonatus and the fourth branch contained S. psittacus and S. frenatus. In conclusion, the obtained results exhibited that the five Scarus species are distinctive ones. In addation, the RAPD patterns are suitable tools to differentiate between fish species.

To study genetic relationship of Cyprinus carpio var. communis, Cyprinus carpio var. specularis and Carassius carassiuscollected from different locations of Kashmir, India, two molecular approaches viz. PCR-RFLP of Cyt b gene and random amplified polymorphic DNA (RAPD) were used. Twenty five samples for each of the species collected from different locations of Kashmir, India were used for this study. Of the 12 random primers used 8 generated the polymorphism, while as four out of 7 restriction enzymes used gave polymorphic restriction patterns. The average proportion of polymorphic markers across restriction endonucleases was 74.4%, whereas random primers showed 82.23% polymorphism. The study showed RAPD marker efficiently helped in inter-and intra-species variation of the species. In contrast PCR-RFLP showed bias in detecting intraspecific variations. However, it was found an efficient tool for inter-specific variation between the species under study. The unweighted pair group method with averages (UPGMA) was used to construct two separate dendrogram for RAPD and PCR-RFLP which resulted in two clusters, one containing C. c. communis and C. c. specularis and the other included the C. carassius species. The Phylogenetic analysis demonstrated that scale carp (C. c. communis) is closest to mirror carp (C. c. specularis) than crucian carp (Crassius carassius). Mean values of haplotype and nucleotide diversity across the species were 0.0200 and 1.0000±0.0113, respectively. Adding more populations with increased sample size of each species will be imperative to draw more definitive conclusions about species characterization within this region.
