Charcoal rot of soybean (Glycine max (L.) Merr.), caused by Macrophomina phaseolina (Tassi) Goid., is an economically important disease worldwide. In recent years, charcoal rot has become a concern for farmers in north-western Argentina. The present work aimed to (1) evaluate disease incidence of charcoal rot; (2) assess morphological and genetic characteristics of M. phaseolina isolates from soybean and other hosts collected in north-western Argentina; and (3) compare genetic diversity between soybean isolates of M. phaseolina from Argentina and the USA. Incidence of charcoal rot evaluated in 11 locations during five growing seasons (2008-2012) was 1%, 1%, 1%, 5-10% and 90%, respectively, indicating a gradual increase over the cropping seasons. Cluster analysis by AFLP of Argentinean isolates exhibited one main group, with isolate Mp56 separated at a genetic distance of 0.70. Four main groups were identified using SSR markers at the same genetic distance, with Mp56 and Mp53 separated from them. There was no clear association between AFLP, SSR profiles, morphological characteristics or host of origin within isolates from Argentina. However, using principal coordinate analysis of SSRs, 22 isolates of M. phaseolina from soybean from north-western Argentina were clearly distinguished from 11 isolates previously collected from two states in the USA. Our results confirmed the increasing importance of charcoal rot in north-western Argentina and identified characteristics of isolates that may be useful for breeding for disease resistance and developing integrated management programmes for charcoal rot in Argentina in the future.Keywords: charcoal rot, disease development, Macrophomina phaseolina, pathogen genetic diversity, SSR markers Résumé: La pourriture noire du soya (Glycine max (L.) Systemic Acquired Resistance (SAR) induced by agrochemicals of chitosan (CH) and salicylic acid (SA) at (25, 50 and 100 ppm) , in addition to a biocontrol agent of T. harzianum (Th) at 4 ×106 were examined against M. phaseolina the causal agent of charcoal rot of sunflower. The results depended on estimation of diseases severity and microsclerotia density in the soil. Thus, the seeds immersion in CH 75 ppm for 6 h., gave the highest and considerable reduction (p=0.05) in disease severity by 48.25% and reduced microsclerotia survived in the soil up to 70%. Application of SA at 50 and 75 ppm proved an obvious reduction of charcoal rot severity by up to 39% and 37% for both concentrations, respectively and not varied with Th. The results also confirmed that CH at 75 ppm revealed significant reduction 40.63% in disease severity and similarized with SA at same concentration. However, the lowest dose of SA at 25 ppm realized the highest reduction of micro sclerotia density by 80.28 % compared to 74.91% when used CH at 75 ppm. For molecular identification of a pathogen Polymerase Chain Reaction (PCR) using ITS4 and ITS5 universal primers were applied to amplify and sequence of DNA for six isolates of M. phaseolina viz., OL901219, OL636051, OL901220, OL901204, OL636050 and OL636053 compared for identity of rDNA sequence according to NCBI GenBank databases by BLAST mode and the results showed the entire similarity ratio reached to 100%

Molecular Identification and Genetic Characterization of Macrophomina phaseolina Strains Causing Pathogenicity on Sunflower and Chickpea

Macrophomina phaseolina is the most devastating pathogen which causes charcoal rot and root rot diseases in various economically important crops. Three strains M. phaseolina 1156, M. phaseolina 1160, and M. phaseolina PCMC/F1 were tested for their virulence on sunflower (Helianthus annuus L.) and chickpea (Cicer arietinum L.). The strains showed high virulence on both hosts with a disease score of 2 on chickpea and sunflower. The strains also increased the hydrogen per oxide (H2O2) content by 1.4- to 1.6-fold in root as well as shoot of chickpea and sunflower. A significant increase in antioxidant enzymes was observed in fungal infected plants which indicated prevalence of oxidative stress during pathogen propagation. The M. phaseolina strains also produced hydrolytic enzymes such as lipase, amylase, and protease with solubilization zone of 5–43 mm, 5–45 mm, and 12–35 mm, respectively. The M. phaseolina strains were identified by 18S rRNA and analyzed for genetic diversity by using random amplified polymorphic DNA (RAPD) markers. The findings based on RAPD markers and 18S rRNA sequence analysis clearly indicate genetic variation among the strains collected from different hosts. The genetically diverse strains were found to be pathogenic to sunflower and chickpea.
