“…Several authors have tried different molecular tools to search for polymorphic markers in coffee. These molecular techniques include RAPDs (Diniz et al 2005;Chaparro et al 2004), ISSRs (Ruas et al 2003), and SSRs (Moncada and McCouch 2004), but most have revealed the low polymorphism of the species and have shown the difficulty of finding polymorphic markers suitable for the construction of the C. arabica molecular map. Here we explored three different molecular tools, Cleaved and Amplified Polymorphic Sequences (CAPS), Single Strand Conformation Polymorphism (SSCP) and Single Nucleotide Polymorphism (SNPs), to identify the best strategy to find polymorphic markers in C. arabica, a crop with a very low genetic diversity.…”

Chaparro et al (2004) used RAPD technique for the analysis of 50 wild and semiwild accessions of C. arabica and concluded that a much larger polymorphism was present in the collection examined than previously reported in others coffee collections.
