Genetic polymorphism of VEGF: Impact on longitudinal change of peritoneal transport and survival of peritoneal dialysis patients

Abstract: We conclude that in peritoneal dialysis patients, the AA genotype of VEGF promoter at -2578 position was associated with progressive increase in peritoneal transport. The CA/AA genotype at -2578 position was also associated with an excess mortality. Our finding also suggests that systemic and local peritoneal VEGF production may be differentially regulated.

“…The CC genotype of VEGF RS3025039 is shown to be associated with higher VEGF levels in the Bahraini population [32]. The CC genotype for VEGF RS699947 was shown to be associated with higher VEGF protein levels in a study on peritoneal dialysis patients [33]. The race distribution in these data was not described and, therefore, may be different from our population.…”

“…Among them, the VEGF -2578C/A has been shown to functionally affect the mRNA level of VEGF. The VEGF -2578C/A polymorphism has been proven to be associated with many diseases, such as lung carcinoma, atherosclerosis, Alzheimer's disease, peritoneal membrane function, graft survival, and breast cancer [41,42,43,44,45,46]. Furthermore, it was reported further that VEGF -2578C/A is associated both with type 1 diabetes and acceleration of its onset [47].…”

“…Anti-VEGF-A antibodies can ameliorate the increase in solute transport, the expression of adhesion molecules [21] , and the anatomical changes induced by peritonitis or bio-incompatible dialysate [11] . Polymorphism of the VEGF-A gene predicts the change in PET during long-term follow-up [22] . In our study, we found that dialysate VEGF-C and IL-6 correlated with PET, but not VEGF-A.…”
