Genetic mapping and annotation of genomic microsatellites isolated from globe artichoke

Abstract: Cynara cardunculus includes three taxa, the globe artichoke (subsp. scolymus L. Hegi), the cultivated cardoon (var. altilis) and their progenitor, the wild cardoon (var. sylvestris). Globe artichoke is an important component of the Mediterranean rural economy, but its improvement through breeding has been rather limited and its genome organization remains largely unexplored. Here, we report the isolation of 61 new microsatellite loci which amplified a total of 208 alleles in a panel of 22 C. cardunculus genoty… Show more

“…An initial set of 93 genomic SSR assays developed in our laboratory (Acquadro et al 2003(Acquadro et al , b, 2009) was used to establish informativeness by amplifying the DNA of the three parents and six plants of each progeny. Each 20 ll PCR contained 10 ng genomic DNA, 10 mM Tris-HCl (pH 8.3), 50 mM KCl, 2.5 mM MgCl 2 , 0.5 U Taq polymerase, 0.2 mM dNTP, 200 nM unlabelled reverse primer and 200 nM IRD700-labelled forward primer.…”

“…Microsatellites (SSRs) are particularly suited to DNA fingerprinting as well as for identifying potential parental genotypes for highly segregating populations development, given their robustness and informativeness. Over the last few years, a substantial set of globe artichoke SSR assays has been developed (Acquadro et al 2003(Acquadro et al , b, 2009, and these have been used to assess genetic diversity Portis et al 2005a, b, c) and to construct genetic linkage maps (Lanteri et al 2006;Portis et al 2009). …”

“…The set of SSRs comprised 247 loci and were assayed using a combination of CELMS (Cynara Enriched Library MicroSatellite; Acquadro et al 2009) and CyEM (Cynara Expressed Microsatellite; Scaglione et al 2009) markers. An initial screen was performed using as template DNA from the two parentals and six of the F 1 progeny, in order to identify those markers which were more informative.…”

“…This can be laborious, time consuming and costly, then, the use of SSR technology depends usually, on the availability of suitable primers. Several SSR markers have been developed for C. cardunculus (Acquadro et al, 2003(Acquadro et al, , 2005a(Acquadro et al, ,b, 2009), so it is not a limiting variable in this species.…”
