Artificial cross between two genetically different populations of JapaneseMisgurnus loach was made to examine the reproductive capacity of the artificial inter-populational hybrid females. Ploidy status and microsatellite genotypes of the eggs laid by these hybrids were inferred from those determined in progenies developed by normal fertilization with haploid loach sperm, induced gynogenesis with UV-irradiated goldfish sperm and/or hybridization with intact goldfish sperm. Some hybrid females laid unreduced diploid eggs genetically identical to the mother. However, these diploid eggs could not develop by spontaneous gynogenesis, but grow to triploid by incorporation of a sperm nucleus. Other hybrid females laid haploid eggs together with diploid eggs and/or various aneuploid and polyploid eggs. Thus, a disruption of normal meiosis occurred in inter-populational hybrid females. The results suggested that the two populations should be so distant as to give rise to atypical formation of unreduced and other unusual eggs in their hybrids.

The purpose of this review is to introduce surrogate production as a new technique for fish-seed production in aquaculture. Surrogate production in fish is a technique used to obtain the gametes of a certain genotype through the gonad of another genotype. It is achieved by inducing germ-line chimerism between different species during early development. Primordial germ cells (PGCs) are a key material of this technique to induce germ-line chimera. In several species, it has been reported that PGCs differentiated from the blastomeres inherited some maternally supplied mRNA located at the terminal regions of the early cleavage furrows. PGCs from donor species (or strains) are isolated and transplanted into host species to induce the germ-line chimera.Four methods for inducing germ-line chimera, namely, blastomere transplantation, blastoderm-graft transplantation, transplantation of PGC from the genital ridge and transplantation visualized PGC with GFP fluorescence, are described. Several problems preventing the successful induction of germ-line chimera in various fish species are mentioned. Surrogate production, however, has the possibility to allow efficient fish-seed production and effective breeding and transfer of biodiversity to an aquaculture strain.Conservation and efficient utilization of genetic resources will be achieved through surrogate production combined with the cryopreservation of PGCs.

Triploidy in the hematology of jundia juveniles (Siluriformes: Heptapteridae)

This study compared the hematological characteristics of diploid and triploid of jundia, Rhamdia quelen juveniles, an important freshwater fish cultured in south Brazil. Hematological morphometry of erythrocytes were determined in blood smears under a light microscope. The blood was used to measure the number of red blood cells (RBC) with a hemocytometer Neubauer chamber, and the numbers of white blood cells (WBC) and thrombocytes that were obtained using an indirect method. The results showed that triploidy increased (p < 0.01) the size and volume of the erythrocytes. Nevertheless, as expected, triploidy decreased (p < 0.01) the number of circulating erythrocytes, leucocytes and trombocytes in the blood of jundia. Moreover differential cell counts were different in diploids and triploids, suggesting that triploidy affects the number of cells and their proportion in blood. Lymphocytes were the most predominant cells in the differential counting of diploid fish (62.5%) while monocytes were predominant in triploid fish (49.6%). These results suggest performance differences between ploidies of jundia, and require future studies to evaluate the potential of triploid jundia in the culture conditions and resistance to infection.Keywords: Rhamdia quelen, triploid, diploid, blood cells. Triploidia na hematologia de juvenis de jundiá (Siluriformes: Heptapteridae)Resumo O presente estudo comparou características hematológicas de juvenis de jundiás Rhamdia quelen diploides e triploides, um importante peixe de água doce cultivado no sul do Brasil. A morfometria hematológica dos eritrócitos foi determinada em extensões sanguíneas sob microscopia óptica comum. O sangue dos animais foi também utilizado para mensurar o número total de eritrócitos em câmera de Neubauer, bem como os números totais de leucócitos e trombócitos pelo método indireto. Os resultados demonstraram que a triploidia aumentou (p < 0,01) o tamanho e volume dos eritrócitos. No entanto, como esperado, a triploidia reduziu (p < 0,01) o número de eritrócitos, leucócitos e trombócitos circulantes do jundiá. Além disto, a contagem diferencial de células sanguíneas foi diferente em diploides e triploides, sugerindo que a triploidia afeta o número das células, bem como sua proporção no sangue. Linfócitos foram as células predominantes na contagem de leucócitos de peixes diploides (62,5%), enquanto os monócitos foram os predominantes nos peixes triploides (49,6%). Estes resultados sugerem desempenhos diferentes entre as ploidias do jundiá e exige estudos futuros para avaliar o potencial dos jundiás triploides sob condições de cultivo e frente à infecção.Palavras-chave: Rhamdia quelen, triploide, diploide, células sanguíneas.
