“…The EcoRI primers were labeled with the fluorescent dye 6=-carboxyfluorescein to enable detection with an ABI Prism 3130 genetic analyzer (Applied Biosystems, LLC, Foster City, CA). Reaction conditions and the thermocycler program used for selective amplification were as described by Terashima et al (48,49). AFLP fragments were detected using the ABI Prism 3130 genetic analyzer, and their sizes were analyzed using Gene Mapper v. 4.0 software (Applied Biosystems).…”

“…In our experiment, all the 19 genotypes were cultivated under the same conditions with the fruiting temperature set at 13 15 C. The strain HmJ2632 was found to grow at a signifi cantly slower rate when compared to the others. Terashima et al (2002) reported that the cultivated L. edodes strains with the same fruiting season type shared the same gene pool. Zhang et al (2007) also observed a similar phenomenon in Chinese cultivated strains of L. edodes.…”

“…Developing and genotyping AFLP markers have been reported in several fungi (Boucias et al, 2000;Majer et al, 1996;Purwantara et al, 2000). The technique has also been applied to genotypic identifi cation in only a few edible mushrooms such as Lentinula edodes (Terashima and Matsumoto, 2004;Terashima et al, 2002) and Pleurotus eryngii (Urbanelli et al, 2007). However, to our best knowledge, there is no report that developed a reliable experimental protocol for genotyping H. marmoreus with AFLP.…”

“…Meanwhile, Vos et al (1995) developed AFLP (amplified fragment length polymorphism) analysis based on selective PCR amplification of genomic DNA restriction fragments, and demonstrated that AFLP analysis could reproducibly detect a large number of polymorphic loci. Furthermore, we previously reported that AFLP analysis reproducibly identified more DNA markers than isozyme and RAPD profiling and that this technique could be used to differentiate major commercial strains of Japanese shiitake (Terashima et al 2002) using freeze-dried mycelia.…”
