Genetic divergence among and within Colletotrichum lindemuthianum races assessed by RAPD

Abstract: Genetic divergence within and among races of was determined using RAPD markers. In addition to the different races of the fungus three isolates of the sexual stage of ( f.sp. ) were included in this study. The band patterns generated using 11 primers produced 133 polymorphic bands. The polymorphic bands were used to determine genetic divergence among and within the pathogen races. The isolates analyzed were divided into six groups with 0.75 relative similarity. Group VI, formed by three isolates of the sexual … Show more

“…Although some authors (Sicard et al 1997;González 1998) have described a tendency of isolates to cluster by region of origin, Mahuku & Riascos (2004) reported no separation of groups according to host genetic pool or geographic region. Similar results have been obtained by Fabre et al (1995), Balardin et al (1997Balardin et al ( , 1999, Talamini et al (2006) and Silva et al (2007).…”

“…In these investigations, races from various countries were evaluated, and when several isolates per race were analyzed they generally clustered in different genetic similarity groups showing the existence of within-race variability. Additionally, Alzate-Marin et al (2001), Talamini et al (2006) and Silva et al (2007) have observed within-race variation in Brazilian isolates. Based on the clustering of similarity estimates for anastomosis, a high number of anastomosis groups were observed within C. lindemuthianum race 65, however, of the 13 isolates investigated only three (LV58, LV61 and LV73) belonged to the same anastomosis group.…”

“…On the other hand, the isolate 4 from the State of Minas Gerais revealed the presence of one SNP in the ITS1 region and one in the ITS2 region, both are different from the SNPs observed in the other isolates. Studies conducted by Talamini et al (2006) noted the presence of molecular variability between and within isolates belonging to race 65 from the State of Minas Gerais.…”

“…The common bean anthracnose is one of the main fungal diseases affecting this crop, especially if the environmental conditions favor the pathogen's development, leading to yield losses of up to 100% when susceptible cultivars are used (Talamini et al, 2006; Damasceno e Silva, Souza, & Ishikawa, 2007 pathogen and is therefore widely distributed in common bean-producing regions (Bianchini, Maringoni, & Carneiro, 2005). Anthracnose occurs more severely in places where relative humidity conditions above 91% and temperatures ranging from 18° and 22°C predominate.…”
