Genetic Characterization of Russian Rapeseed Collection and Association Mapping of Novel Loci Affecting Glucosinolate Content

Abstract: Rapeseed is the second most common oilseed crop worldwide. While the start of rapeseed breeding in Russia dates back to the middle of the 20th century, its widespread cultivation began only recently. In contrast to the world’s rapeseed genetic variation, the genetic composition of Russian rapeseed lines remained unexplored. We have addressed this question by performing genome-wide genotyping of 90 advanced rapeseed accessions provided by the All-Russian Research Institute of Oil Crops (VNIIMK). Genome-wide gen… Show more

“…To perform genotyping, half-seeds of sunflower lines left after cotyledon separation were germinated in rolls of filter paper, after which DNA was extracted from the cotyledon leaves using NucleoSpin Plant II kit (Macherey-Nagel) according to the manufacturer’s manual. GBS library was prepared according to the modified protocol [ 24 ] as described previously [ 25 , 26 ]. GBS library sequencing was performed in Illumina HiSeq 4000 with single-end reads with a length of 150 bp.…”

“…Wei et al (2019) carried out genome-wide resequencing in a population of diverse B. napus accessions with different phenotypes, and identified several loci, i.e., orthologs of MYB28, MYB34, and AOP3, that affecting seed GSL level (Wei et al 2019). Similarly, Gubaev et al (2020) applied the genotyping-by-sequencing approach in 90 advanced rapeseed accessions and found new candidate genes that potentially contribute to the control of GSL content, such as genes encoding the histone acetyltransferase HAC1 and BES1/ BZR1 homolog protein 4-like (Gubaev et al 2020).…”
