Genetic association of glutathione peroxidase-1 with coronary artery calcification in type 2 diabetes: a case control study with multi-slice computed tomography

Abstract: Background: Although oxidative stress by accumulation of reactive oxygen species (ROS) in diabetes has become evident, it remains unclear what genes, involved in redox balance, would determine susceptibility for development of atherosclerosis in diabetes. This study evaluated the effect of genetic polymorphism of enzymes producing or responsible for reducing ROS on coronary artery calcification in type 2 diabetes (T2D).

“…The 8-isoprostanes are formed during the free radical-mediated oxidation of arachidonic acid (12), which is ubiquitously distributed in the cell membrane, and 8-isoprostanes have been reported as a marker of whole body oxidative stress (13). Our results indicate that the whole body oxidative stress was significantly increased in patients with type 2 diabetes, and this was associated with a higher CACS, as reported previously in Western subjects (4) and in Japanese subjects with a gene associated with susceptibility to oxidative stress (14).…”

“…In both T1DM and T2DM, the Val/Val genotype is associated with an increased risk of diabetic nephropathy (Mollsten et al, 2007;Nomiyama et al, 2003). Another study in 91 T2DM patients found no association between SOD2 genotype and coronary artery calcium score (Nemoto et al, 2007). The results of these studies are difficult to compare due to the different co-morbidities investigated.…”

“…Decreased GPX-1 activity is associated with increased risk of atherosclerosis (Espinola-Klein et al 2007). The GPX1 gene polymorphisms associated with decreased GPX1 enzymatic activity have been associated with increased risk of atherosclerosis (Hamanishi et al 2004;Nemoto et al 2007;Tang et al 2008;Winter et al 2003). Glutathione stransferase (GST) is another enzyme that plays a key role in cellular antioxidant defence mechanisms.…”
