Association of Coronary Artery Calcification with MDA-LDL-C/LDL-C and Urinary 8-Isoprostane in Japanese Patients with Type 2 Diabetes

Abstract: Objective Oxidative stress has been implicated in the development of coronary artery calcification (CAC). However, there are few reports on this issue in Japanese patients with diabetes. In this study, we examined the association of the CAC score (CACS) with oxidative stress markers. Methods The study subjects were 163 Japanese patients with type 2 diabetes (75 men and 88 women). The CACS (Agatston unit: AU) was measured by multi-detector computed tomography (MDCT), and the oxidative stress markers, such as th… Show more

“…IMT of the carotid arteries was measured using ultrasound diagnostic equipment (LOGIQ 500, GE Yokogawa Medical Systems Corp., Hino, Tokyo, Japan) with an electrical linear transducer (mid-frequency of 7.5 MHz). The common carotid artery, carotid bulb, and portions of the internal and external carotid arteries on both sides were scanned with the subject in the supine position 6) . We defined the max IMT as the thickest portion detected in the scanned regions.…”

“…CACS measurement has also been regarded as being clinically useful for evaluating coronary atherosclerosis in Japan 5) where the incidence of coronary heart disease is much lower than in Western countries. In a previous study, we showed CACS to be positively associated with biological markers of oxidative stress 6) and to predict the morbidity of CVD in Japanese subjects with type 2 diabetes 7) .…”

“…Oxidative stress markers, such as urinary 8‐isoprostane and 8‐hydroxydeoxyguanosine (8‐OHdG) and serum malondialdehyde‐LDL cholesterol (MDA‐LDL‐C), were measured by SRL, Inc. (Tokyo, Japan). Hepatic steatosis was defined as a liver to spleen density ratio below 0.9, based on plain abdominal CT .…”

“…Serum MDA-LDL levels have been positively correlated with carotid intima-media thickness [ 9 , 12 ]. Moreover, it was shown that the MDA-LDL-to-LDL-cholesterol (LDL-C) ratio (MDA-LDL/LDL-C) was a more useful predictor of coronary artery calcification than the MDA-LDL level alone [ 11 , 13 ]. Kurobe et al revealed that the vascular protective effects of ezetimibe, an inhibitor of cholesterol intestinal absorption, were correlated with decreased values of MDA-LDL and MDA-LDL/LDL-C but not with those of LDL-C [ 14 ].…”
