By means of the “prosome” analysis technique, by which chromosomes are studied at early metaphase and even at prophase, an improved resolution of the G‐band pattern has been achieved. The 2 chromosome regions of especial significance in the (8; 14) translocation of Burkitt's lymphoma, 8q24 and 14q32, which in the Pans karyotype were represented as uniformly negative, in prosome analysis resolved into 5 and 6 subbands respectively. The (8; 14) translocation was shown to be reciprocal and the breaking points could be established in both chromosomes with more precision than earlier. It was noticed that the segment of No. 8, translocated onto No. 14, exhibited somewhat different morphologic features in translocated than in normal position. The concept of local prosomisation was introduced and discussed.

A man and two of his three children carried an abnormally short chromosome 22 resembling the Philadelphia chromosome (Ph1). Giemsa banding showed that the abnormal chromosome resulted from a translocation t(11;22) (q25;q13). The breakpoint on chromosome 22 was at the q12/q13 band interface compared with the breakpoint of Ph1 at the q11/q12 band interface. The absence of leukaemia or haematological disorder in members of this family suggests that the critical genetic site on chromosome 22 concerned with abnormal myeloid cell proliferation in human leukaemia is contained in the 22q12 band.

The R-banding pattern of the chromosomes of 31 patients hospitalized in the Hematologic Clinic for myeloid leukemia were studied before chemotherapy. This analysis permitted identification of one unusual 3-chromosome rearrangement t(3;9;22) in addition to 25 classic forms of (22q-;9q+) translocation accompanied by the specific Ph' chromosome in chronic granulocytic leukemia patients, independent of the blastic course of the disease. During blastic crisis observed in 6 patients, extra 8 and 10 chromosomes, monosomy for chromosome 17, isochromosomes 17q, translocation (12q;13q), and additional Ph' were noted. The nonrandomness of these findings is determined from results published by other authors. Their significance for the cellular phenotype is presently unknown.
