Genes for the tumor necrosis factors alpha and beta are linked to the human major histocompatibility complex.

Abstract: The human major histocompatibility complex (MHC) includes the closely linked genes for the tumor necrosis factors a and (3. Their location is within the chromosomal segment between HLA-DR and HLA-A or centromeric of HLA-DP. This assignment is based on Southern blot analysis of a number of different MHC deletion mutants and is corroborated by chromosome in situ hybridization.

“…The genes for both TNF-alpha and TNF-beta are separated by about I kb of DNA on chromosome 6 within the major histocompatibility complex (Nedwin et al, 1985a;Spies et al, 1986). The expression of TNF mRNA encodes a precursor of 233 amino acids that is processed to a mature non-glycosylated protein of 17,300 Da containing a single disulphide linkage (Wang et al, 1985).…”

“…TNF-a, a cytokine with pleiotropic effects, satisfies the four candidate criteria alluded to above (for reviews, see Jaattela, 1991;Vilcek and Lee, 1991). TNF-a maps to the D region of the H-2 complex (Muller et al, 1987); it also maps to the human HLA region on the short arm of chromosome 6 (Spies et al, 1986). The TNF-a gene is polymorphic and this polymorphism is associated with differences in the level of TNF-a protein expression (Jacob and McDevitt, 1988) and with H-2 haplotype differences (Freund et al, 1992).…”

“…(Spies et al, 1986;Carroll et al, 1987;Ziegler et al, 1991). The primary transcript of human TNF-a consists of 2,762 bp which has four exons and three introns (Nedwin et al, 1985;Nedospasov et al, 1986).…”
