Generation of induced pluripotent stem cells from primary chronic myelogenous leukemia patient samples

Abstract: Induced pluripotent stem cells (iPSCs)can be generated by the expression of defined transcription factors not only from normal tissue, but also from malignant cells. Cancer-derived iPSCs are expected to provide a novel experimental opportunity to establish the disease model. We generated iPSCs from imatinib-sensitive chronic myelogenous leukemia (CML) patient samples. Remarkably, the CML-iPSCs were resistant to imatinib although they consistently

This cell population may therefore be useful to interrogate the role of stem cells in CML and the mechanisms underlying the development of resistance [72] . iPSCs and pancreatic ductal adenocarcinoma iPSCs have also been used to study pancreatic ductal adenocarcinoma (PDAC), which currently has no suitable model.…”

“…Disease Modeling Using Patient-Specific iPSCs To date, several patient-specific iPSC lines have been generated from patients with a wide variety of mainly monogenetic, early-onset diseases such as neurologic disorders, 44,45) heart disease, 46,47) metabolic disease, 48) hematologic disorders, 49) mitochondrial disease, 50) chromosomal abnormalities, 51) telomere disease, 52,53) sensory organ disorder, 54) and storage disease.…”

“…Since the invention of this method, iPSCs have been established from a variety of somatic cells not only for regenerative medicine but also for studies of the pathogenesis of inherited genetic disease 2-5 or neoplasms [6][7][8][9] . In term of the establishment of iPSCs from blood cells, the T cells that were derived from antigen-specific CD8 + T cells in an HIV-1-infected patient 10 , or from mature cytotoxic T cells that were specific for the melanoma epitope MART-1 11 , were reprogramed into iPSC, and were then re-differentiated into CD8…”
