Gene number in species of Astereae that have different chromosome numbers

Abstract: Differencesin the gametic chromosome numbers (n = 4, 5, 9) ofspecies.in the Astereae tribe ofthe Compositae have been variously .interpreted. One hypothesis proposes that n = 9 was the original base number ofthe group and that the lower numbers resulted from aneuploid reduction. The alternative hypothesis asserts that the ancestral base number was n = 4 or n = 5 and that species in which n = 9 are allotetraploids derived by hybridization between taxa with the lower numbers. Electrophoretic analysis of 17 enzy… Show more

“…Two to six fresh young leaflets from each plant were crushed in an ELISA box, using a glass rod after the addition of 4 drops of extraction buffer (Gottlieb 1981 slightly modified by Gaur & Slinkard 1990 a,b) . Extracts were absorbed into filter paper wicks (Whateman no .…”

“…A Tris-EDTA-maleic acid system, pH 7.4 (Brown et al, 1978) was used for GP, LAP and NADHD. LiOH-borate, pH 8.3, was used for ACPH, ALD, CAT, EST and GPI; and 0.02 M histidine-citrate, pH 7.0, was used for the other enzymes (Gottlieb, 1981).…”

“…This problem is to some extent circumvented with biochemical markers such as isozyme loci, where the in vivo function of the genetic products is often clearer, resulting in an increased probability of correctly identifying homologous genes. Because of this, isozyme gene number has proven to be a valuable source of evidence regarding the ploidy level of various plant taxa (Gottlieb 1981(Gottlieb , 1982.…”
