Gene Functional Analysis Using Protoplast Transient Assays

Abstract: The protoplast transient assay system has been widely used for rapid functional analyses of genes using cellular and biochemical approaches. This system has been increasingly employed for functional genetic studies using double-stranded (ds) RNA interference (RNAi). Here, we describe a modified procedure for the isolation of protoplasts from leaf mesophyll cells of 14-day-old Arabidopsis thaliana. This modification significantly simplifies and speeds up functional studies without compromising the yield and the… Show more

“…Seeds of the model grass species B. distachyon (Bd21‐3) were obtained from the French National Institute for Agricultural Research (INRA). Seeds were grown hydroponically following the protocol of Jung, Yan, Zhai, and Vatamaniuk (2015). Briefly, seeds were soaked in water for 2 hr to soften the lamella and palea which were then removed using forceps.…”

“…The hydroponics set up consisted of two nested disposable cups that contained a foam disc cut to size with three slots cut to hold plants (see Figure S1). Each cup was filled with approximately 330 ml of full strength nutrient solution as per Jung et al (2015) except that Fe(III) EDTA concentration was doubled due to iron deficiencies detected in trial plants.…”

“…Protoplasts were isolated from 2‐week‐old seedlings grown on 1/2 MS plates as described previously (Jung, Yan, Zhai, & Vatamaniuk, ). To create fusion proteins with the EGFP tag, full‐length cDNA was PCR‐amplified and cloned into the pSAT6‐EGFP vector (Tzfira et al, ).…”

“…A step-by-step protocol for protoplast isolation from leaf mesophyll cells of the Bd21-3 inbred line has been published by Jung et al (2015). We present the optimized protocol for the isolation of protoplasts from B. distachyon Bd21.…”
