Gene for parathyroid hormone-like peptide is on mouse chromosome 6

Abstract: The single-copy parathyroid hormone-like peptide gene (Pthlh) was assigned to mouse chromosome 6 using a rat PTHLH cDNA as hybridization probe in Southern blot analysis of DNAs isolated from a panel of mouse × Chinese hamster cell hybrids. The mouse parathyroid hormone gene (Pth) has previously been assigned to mouse chromosome 7 and the PTHLH nd PTH genes have also been shown to be on different chromosomes in human and rat. Therefore, despite significant amino-terminal sequence homology between the PTHLH and … Show more

“…For standardization of DNA PCRs, amplification was performed in parallel (again in triplicates) on a 90-bp fragment of the single-copy mouse gene (pthlh) for parathyroid hormone-related protein (NM008970) (7,26). The relative copy numbers of the retroviral gene and of the pthlh gene were calculated by use of separate standard curves (serial dilution [0.1 to 10 ng] of SELH or LM DNA on the same plate), and standardization was carried out by division of the relative number of retroviral genes by the relative number of pthlh amplicons.…”

“…The mouse Pthlh has been assigned to chromosome 6 using somatic cell hybrids (Hendy et al" 1990). The present study now localizes Pthlh to band F-G by in situ hybridization.…”

“…The single copy PTHLH gene has been assigned to human chromosome region 12p 12.1 -?• p 11.2 (Mangin et al, !988;Suva et al, 1989; mouse chromosome 6 (Hendy et al" 1990), and rat chromosome 4 (Szpirer et al" 1991). The rodent chromosomes contain syntenic groups conserved on human chromosome 12p.…”

“…The cDNA was found to encode a protein of 177 amino acids (molecular weight 16kDa), which consisted of a precursor sequence of 36 amino acids and a mature peptide of 141 amino acids. Analysis of genomic DNA from four species revealed a pattern compatible with a single-copy gene, and the PTHrP gene was mapped to chromosome 12p12.1-pl 1.2 in man (Mangin et al 1988) and to chromosome 6 in the mouse (Hendy et al 1990). The human PTHrP gene, which is 14kb in size and contains 7 exons (Mangin et al 1989;Yasuda et al 1989), was found to be normally expressed in keratinocytes and other tissues which include the parathyroids, adrenals, thyroid, pituitary, fetal liver and lactating mammary glands.…”
