Gender-related differences observed among immune cells in synovial fluid in knee osteoarthritis

Abstract: Based on the immune cell composition of SFs, data mining analysis revealed distinct phenotypes (monocyte- and lymphocyte-predominant) within each gender group. This first study on the cellular complexity of SFs in KOA showed marked differences between male and female patients. The findings give a rational starting point for patient stratification according to their phenotypes, as is required for phenotype-specific treatment strategies.

“…Cells in whole blood were stained with optimal concentrations of monoclonal antibody combinations directed against the following surface antigens: CD45-PerCP/Cy5.5, CD5-PE, CD19-APC/Cy7, CD54-FITC, CD62L-APC, CD49d-PE/Cy7, CD183-FITC (CXCR3), CD184-APC (CXCR4), CD185-FITC (CXCR5), CD197-PE/Cy7 (CCR7), CD195-PE/Cy7 (CCR5), CCR10-APC (all BioLegend), as reported previously [ 14 , 15 ]. Isotype-matched antibodies were used as negative controls.…”

“…Unlike their female counterparts, inguinal lymph nodes of adult male mice had no significant differences in macrophage numbers after 1 or 2 weeks of repetitive loading compared to the contralateral control ( Figure 2B). The gender dependency could be attributed to higher prevalence of macrophages in women than men with OA (14). Furthermore, within female mice, this response was age dependent.…”

“…They also induce monocyte/macrophage-derived osteoclast activity via RANK-L secretion resulting in enhanced bone resorption ( Yoshitomi, 2019 ). It is interesting to note given the above pro-inflammatory properties that the presence and activity levels of these tissue-resident macrophages significantly correlates with advanced stages of OA and poorer clinical outcome scores ( Kriegova et al, 2018 ; Gomez-Aristizabal et al, 2019 ).…”
